β-纤维蛋白原基因-455G/A多态性与中青年缺血性脑卒中的相关研究

摘　　要：目的探讨血浆纤维蛋白原(fibrinogen,Fg)水平,β-纤维蛋白原-455 G/A(β-Fg-455G/A)基因多态性与中青年缺血性脑卒中的关系。方法应用聚合酶链反应-限制性片断长度多态性方法(PCR-RFLP)对101例中青年(年龄<55岁)急性缺血性脑卒中患者和96例中青年健康对照者的基因多态性进行研究,并测定血浆Fg水平。结果血浆Fg水平病例组和对照组分别为(246.8±53.0)mg/dL和(228.6±24.3)mg/dL,差别有统计学意义(P<0.01);病例组及对照组均有AA+GA型血浆Fg水平高于GG型,差别有统计学意义(P<0.05);各基因型频率和等位基因频率两组间均无统计学意义(P>0.05)。结论血浆Fg水平增高是中青年缺血性脑卒中的危险因素,未发现β-Fg-455G/A基因多态性与中青年缺血性脑卒中相关。Objective To study the relationship between plasma fibrinogen leve, beta- fibrinogen gene - 455G/A polymorphism,and ischemic stroke in mid- adults and young adults. Methods Polymerase chain reaction and restricted fragment length polymorphism （PCR - RFLP） method was applied to determine the polymorphism of beta - fibrinogen gene - 455G/A in 101 patients with ischemic stroke and 96 controls. And plasma fibrinogen level was determined as well. Results The plasma fibrinogen level showed significant difference between the patient and control groups （246.8 ± 53.0mg/dL vs 228.6 ± 24.3mg/dL, P 〈 0.01 ）. No significant difference was found in frequencies of genotypes and alleles between two groups （ P 〉 0.05）. Conclusion Plasma fibrinogen level was a risk factor for ischemic stroke, the association between β- Fg- 455G/A polymorphism and the incidence of ischemic stroke in mid- adults and young adults was not observed.
