摘  要:目的探讨海马局部移植人骨髓间充质干细胞(hMSC)对癫痫持续状态(SE)后大鼠视空间记忆功能的影响。方法雄性SD大鼠随机分为hMSC移植组、人成纤维细胞组、磷酸盐缓冲液(PBS)组、假手术组和正常对照组,采用氯化锂-匹罗卡品诱导SE模型。SE 3 d后通过立体定位方法分别向海马CA1区注射hMSC(1.5×107个/ml)、人成纤维细胞(1.5×107个/ml)和PBS各3μl,假手术组不进行立体定向注射。2周后部分大鼠行灌流取脑组织,MAB1281免疫组化染色观察示踪细胞。剩余大鼠在4周后进行水迷宫实验。结果免疫组化染色显示仅在hMSC移植组见到CA1区有染棕黄色的细胞,而人成纤维细胞组、PBS组和假手术组均未见到染色阳性细胞。水迷宫试验显示hMSC移植组的逃逸潜伏期短于人成纤维细胞组和PBS组,仅略长于假手术组,虽与正常对照组比较差异有统计学意义(P<0.05),但除正常对照组以外,其余4组组间两两比较差异均无统计学意义(P>0.05)。结论hMSC可在SE大鼠海马CA1区局部存活,移植后4周未发现hMSC能明显改善SE大鼠的视空间记忆功能。Objective To investigate the influence of intrahippocampal transplantation of human mensenchymal stem cell (hMSC) to visual-spatial memory in rats following status epilepticus (SE).Methods The hMSCs were cultured in vivo.Healthy male SD rats were randomly admitted into 5 groups: control,hMSC,fibroblast cell,PBS and sham groups.All rats except rats in the control group were induced into SE with an injection of Lithium and Pilocarpine.Three days later, three microliters of hMSC (1.5×10^7/ml),fibroblast cell(1.5×10^7/ml) and PBS were given to the CA1 region of the hippocampus of the surviving rats in hMSC,fibroblast cell and PBS group respectively through microsyring in a stereostatic frame.MAB1281 immune-staining was performed to detect transplanted cells 2 weeks later.The morris water maze test was used to assess spatial learning and memory 4 weeks later.Results Brown cells were detected only in the CA1 region of hipocampus of rats in the hMSC group.The escape tendency of rat in the hMSC group was shortened than those in the fibroblast cell and PBS groups,and a little longer than that in the sham group.There was a significant difference in escape tendency between the rats in the hMSC and normal control groups,but no significant difference among rats in hMSC group,fibroblast cell group,PBS group and sham group.Conclusion hMSC can survive in the CA1 region of hippocampus of rats.The visual-spatial memory is not improved in rats following status epilepticus after transplanted with hMSC.
