摘 要:目的了解湖南省狂犬病毒的分子流行病学特征以及其与疫苗株的差异。方法对4株湖南狂犬病病毒P和M基因同时进行了RT-PCR扩增、克隆和测序。结果同源性分析表明,4株湖南街毒P和M基因核苷酸同源性分别为97.3%~99.4%和98.4%~99.8%;与其它I型狂犬病病毒和疫苗株核苷酸同源性比较,P基因分别为78.4%~90.2%和81.8%~86.8%;M基因的分别为81.8%~92.1%和84.7%~90.6%。4株野毒P和M基因氨基酸同源性分别为98.0%~99.3%和98.5%~99.0%。结论4株病毒P基因和M基因的核苷酸序列及推导的氨基酸序列与我国现用的人用及兽用疫苗株存在一定差异。进化关系表明,4株病毒均为基因I型狂犬病毒;与我国宁夏分离株最近;与其它I型毒株分离自菲律宾和泰国等东南亚的毒株亲缘关系较近;而与翼手目的毒株关系最远。Objective To better understand the current rabies epidemics and genetic characteristics of rabies viruses with the vaccine strain in China. Methods The phosphoprotein gene and matrix protein gene of four rabies virus strains isolated from Hunan by reverse transcription - polymerase chain reaction ( RT-PCR) , respectively were amplified. The cDNA frag- ments were cloned and then sequenced. Results Phylogenetic analysis indicate that the strains belonged to classic rabies virus, and shared the higher degree of homology, with 97.3%-99.4% nucleotide identity and 98.0%-99.3% amino acid identity for P gene, 98.4%-99.8% nucleotide identity and 98.5%-99.0% amino acid identity for M gene, respectively. The nucleotide homology between other classic rabies virus strains and vaccine strains ranged from 78.4% to 90.2% and 81.8%-86.8% in Pgene, and 81.8%-92.1% and 84.7%-90.6% in Mgene , respectively. Conclusion Compared with the classic rabies virus, the strains shared the higher degree of homology with the street viruses obtained from Thailand and Philippines than other known strains especially that rabies virus isolated from Chiroptera in America. Further comparing the deduced amino acid sequences showed the strains were more closely related to the vaccine strain CTN than to the other vaccine strains (3aG, PV, PM and ERA). The most important is that both P and M genes diverged from the current vaccine strains used in China at either the nucleotide or the amino acid level.
