摘  要:目的蛋白质组学分析鉴定与肿瘤细胞共培养T细胞的表达差异蛋白。方法双向电泳分离有/无肿瘤细胞共培养的T细胞蛋白,PDQuest软件分析筛选肿瘤细胞诱导表达异常的蛋白点,用基质辅助激光解析电离飞行时间质谱(MAL-DI-TOF MS)对酶解消化样品进行肽质量指纹图谱(PMF)分析,结合数据库搜索鉴定蛋白质。结果PDQuest分析有/无肿瘤细胞共培养的T细胞蛋白双向电泳图谱,70余个蛋白点表达水平差异显著,其中与肿瘤细胞共培养的T细胞有一明显表达下调的蛋白点,质谱分析结合数据库检索表明该蛋白点为腺苷脱氨酶。结论肿瘤细胞诱导T细胞下调表达腺苷脱氨酶。Objective To identified protein differential expression in T cell cocuhured with tumor cells. Methods The whole proteins of Jurkat T cells cocuhured without or with tumor cells were analyzed by two-dimensional electrophoresis (2D-E) whose gels were stained with Coomassie Brilliant Blue R-250. Images were digitalized using GS-800 image scanner and analyzed by PDQuest 2-D analysis software. Gel spots excised from the stained gel were in-gel digested with uypsin and analyzed with MALDI-TOF MS. Results The protein expression levels of more than seventy spots were up- or down- regulated in Jurkat T cells cocuhured with tumor cells. One protein spot of down-regulated expression was identified as adenosine deaminase using MALDI-TOF MS and database searching. Conclusion The expression of adenosine deaminase is down-regulated in Jurkat T cells cocuhured with tumor cells.
