摘 要:目的探讨肝泰对实验性脂肪肝的保护作用及其可能机制。方法SD雄性大鼠53只,随机分为4组,即模型组(n=15)、肝泰组(n=15)、易善复对照组(n=15)?正常对照组(n=8)。利用高脂饮食建立大鼠脂肪肝模型,肝泰组给予肝泰(755mg/kg)干预,易善复对照组给予易善复(550mg/kg)干预,10周后处死所有大鼠。采用光镜及电镜观察肝脏组织学改变,检测大鼠血清总胆固醇(TC)?甘油三酯(TG)?高密度脂蛋白-胆固醇(HDL-C)、肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)及免疫组化法检测肿瘤坏死因子(TNF-α)在肝组织中表达。结果病理所见:模型组出现重度水变性和脂肪变性及点、灶状坏死;肝泰组未见重度水变性和脂肪变性,可见极少数点状坏死。肝泰组肝组织TNF-α与模型组肝组织TNF-α比较,肝泰组TNF-α蛋白含量显著减少(P<0.05)。肝泰组血浆TC和肝匀浆GSH与模型组比较,肝泰组血浆TC较模型组显著减低,肝泰组GSH比模型组显著升高,均有显著性差异(P<0.05)。结论肝泰对实验大鼠脂肪肝有保护作用。Objective To explore the effects of Gantai and its possible mechanisms on preventing fatty liver induced by high-fat feeding. Methods Fifty-three male SD rats were randomly divided into four groups: the normal control group (n=8), the model group (n=15), the Gantai group (n=15) and the Yishanfu control group (n=15).The model of the fatty livers was induced by high-fat feeding. The rats in Gantai group were treated with Gantai (755 mg/ kg). The rats in the Yishanfu control group were treated with Yishanfu (550 mg/kg). All animals were sacrificed at the end of ten weeks following the treatment. Blood, lipids, liver MDA, SOD and GSH were measured. The expression of tumor necrosis factor alpha (TNF-α) was detected by immunohistological staining. Liver histologies from all the groups were analyzed by HE staining and electron microscope. Results The liver histologic examination showed moderate to se- vere steatosis, lobular inflammatory, cell infiltration and necrosis in the model group. Compared with the model group no signs of severe steatosis and fatty degeneration and very few spotty necrosis was observed in the Gantai group. The serum total cholesterol (TC) and the expression of tumor necrosis factor alpha (TNF-α) in the Gantai group were significantly reduced compared with the model group (P 〈 0.05 ). GSH of the liver homogenate in the Gantai group was statistically higher than that in the model group (P 〈0.05). Conclusion Gantai can prevent the formation of the fatty liver.
