摘 要:目的探讨硫酸镍对体外培养的正常人晶状体上皮细胞(LECs)的抑制作用。方法四甲基偶氮唑蓝法测定不同质量浓度的硫酸镍(100、200、300、400、500μg/mL)对人LECs增生的抑制作用,观察时间-效应关系;透射电镜观察细胞的超微结构改变。结果不同质量浓度的硫酸镍作用24h、48h后,LECs的增生抑制率随质量浓度的增加而增高,呈明显的质量浓度-效应关系(r=0.964)。作用24h后LECs的半数抑制质量浓度(IC50)为162.71μg/mL;作用48h后为129.31μg/mL;电子显微镜下观察到细胞核染色质凝集、固缩,核碎裂,大量凋亡小体出现等典型的细胞凋亡的形态学改变。结论硫酸镍对人LECs的增生具有明显的抑制作用,呈明显的质量浓度-效应关系,并能诱导人LECs凋亡。Objective Epidermal study showed that the incidence of cataract is obviously increased in niekel-contact populations,and its mechanism is unclear up to now. We tried to investigate the effect of niekel sulfate on in vitro cultured human lens epithelial ceils. Methods The normal human lens epithelial cell lines were cultured and passaged in DEME medium containing 20% fatal bovine serum and low concentration of glucose. Passaged ceils were divided into blank control group( LECs + DMEM) , negative control group( LECs + DMEM + DMSO) and different concentrations of niekel sulfate treated groups ( 100, 200,300,400,500μg/mL). The inhibitory action of different levels of niekel sulfate on human lens epithelial ceils was detected using methyl thiazolyl tetrazolium(MTT). The ultramicrostructure of cultured ceils was examined under the transmission electron microscope. Results In 24 hours and 48 hours after addition of niekel sulfate,the inhibitory rate was obviously increased and the A value of cultured LECs was significantly reduced with the elevation of concentration of niekel sulfate( 24 hours : F = 39. 496, P = 0. 008 ; 48 hours : F = 17. 547, P = 0. 025 ). Twenty-four hours later, inhibiting concentration of half the number of niekel sulfate was 162.71 μg/mL and that in 48 hours was 129. 31 μg/mL. The typical apoptotic ceils were seen under the electron microscope after addition of different concentrations of niekel sulfate, such as nuclear chromatic agglutination, pyknosis, nuclear fragmentation and lots of apoptotic bodies. Conclusion Niekel sulfate has the inhibitory effect and apoptosis-inducing action on the proliferation of cultured human lens epithelial ceils.
