摘 要:目的:探讨COX-2和VEGF在肝癌多细胞球体(MCS)中的表达与肝癌群集耐药的关系.方法:以liquidoverlay技术获得肝癌HepG2多细胞球体为模型,采用透射电镜和扫描电镜比较单层细胞与MCS的形态学差异;采用MTT法和流式细胞技术比较ADM和5-FU对单层细胞和MCS的生长抑制率和细胞凋亡率;采用免疫组化和图像分析技术比较COX-2和VEGF在单层细胞和多细胞球体表达的差异.结果:与单层细胞相比,多细胞球体细胞间黏附广泛而紧密,可见中间连接和桥粒连接.不同浓度的ADM和5-FU作用48h,MCS细胞的生长抑制率和细胞凋亡率均低于单层细胞,差异有统计学意义(P<0.05).免疫组化结果显示,COX-2和VEGF在MCS上高表达(P<0.05).结论:肝癌MCS具有群集耐药性,可能与其高表达COX-2和VEGF有关.AIM: To examine the expressions of COX-2 and VEGF in hepatocellular carcinoma muhicellular spheroids (MCS) and their relationship with multicellular resistance (MCR). METHODS: Hepatocellular carcinoma HepG2 MCS were obtained by liquid overlay technique and observed by transmission electron microscope and scanning electron microscope. After treatment with ADM and 5-FU for 48 h, cell growth inhibition rate and cell apoptosis rate of monolayer cells and MCS were analyzed by MTr assay and flow cytometry. The expressions of COX-2 and VEGF in monolayer cells and MCS were examined by immunohistochemical method and image analysis. RESULTS: Compared with monolayer cells, MCS had more extensive and compact cell adhesion and intermediate junction and desmosome were found within them. Treated with ADM and 5-FU for 48 h, cell growth inhibition rate and cell apoptosis rate were significantly lower in MCS than in monolayer cells ( P 〈 0. 05 ). The immunohistochemical results showed COX-2 and VEGF expressions were higher in MCS than in monolayer cells ( P 〈 0.05 ). CONCLU- SION : Hepatoeellular carcinoma MCS has muhicellular resistance and the possible mechanisms may be associated with the upregulation of COX-2 and VEGF
