摘  要:目的:研究紧密连接蛋白occludin,ZO-1在豚鼠耳蜗外侧壁血管纹中的表达.方法:成年健康杂色豚鼠,耳廓反射灵敏,分别取耳蜗、脑、肺三组组织,免疫组织化学法及Western Blot研究紧密连接蛋白occludin,ZO-1在耳蜗外侧壁血管纹中的表达,以脑组、肺组为阳性对照.结果:光镜下可见豚鼠耳蜗外侧壁血管纹毛细血管内皮细胞等边缘细胞中均强阳性颗粒表达,脑组、肺组毛细血管内皮细胞内壁也均有明显阳性表达;Western Blot下occludin,ZO-1在耳蜗组中高表达,并且明显高于脑组与肺组.结论:occludin,ZO-1在豚鼠耳蜗外侧壁血管纹紧密连接中的高浓度表达,与维持血迷路屏障结构、功能密切相关.AIM: To investigate the expressions of occludin and ZO-1 in the lateral wall stria vascularis of the guinea pig cochlea. METHODS: Healthy, pigmented, adult guinea pigs of both sexes with intact Preyer's reflex were divided into 3 groups ( coch- lea group,brain group and lung group). The immunohistochemistry and Western Blot were used to detect the expressions of occludin and ZO-1 in the lateral wall stria vascularis of the guinea pig cochlea, as compared to brain and lung. RESULTS: The light microscope demonstrated occludin and ZO-1 presence in the tight junctions of stria vascularis endothelial cells and marginal cells, as well as in the brain and lung. Western Blot analysis revealed strong expressions of occludin and ZO-1 in strial tissue. In addition, the expressions were obviously higher than these in the brain group and lung group. CONCLUSION: The high expressions of occludin and ZO-1 in the tight junctions of stria vascularis in guinea pigs has a relationship with the maintenance of the structure and function of blood-labyrinth barrier.
