摘  要:目的筛选新的肿瘤/睾丸抗原相关基因,为肿瘤的诊断和治疗提供理论依据。方法利用文献报道的大规模平行信号测序技术获得的人类33种正常组织的基因表达谱,经生物信息学分析筛选出睾丸富集表达基因。利用逆转录聚合酶链反应(RT-PCR)技术,在15种人类正常组织中对候选基因进行验证,最后利用荧光定量PCR技术对睾丸特异基因在结肠癌组织和癌旁组织中的表达差异进行分析。结果在15种人类正常组织中,PRM2,PRM1,TNP1,AKAP4,TCP11为睾丸特异性表达,在其它组织中均无表达。其中PRM1基因在50%(6/12)的结肠癌组织中有明显的上调(>2倍)。其它基因在结肠癌中表达与癌旁组织比较无统计学差异。结论PRM1可能是一个新的CT基因,结果显示PRM1基因可能会在结肠癌的分子诊断或者免疫治疗中起到重要作用。Objective To identify novel cancer/testis antigen genes (CT genes). Methods First,to explore the enriched genes expressed in human testis tissue,Analyzed the published gene expression profile of 33 human normal tissues derived from massively parallel signature sequencing (MPSS) method. Second,Validated the expression of testis-enriched genes in 15 human normal tissues through reverse transcription polymerase chain reaction (RT-PCR) to candidate testis-specific expression genes. And then investigated the expression of candidate testis-speclflc genes in 12 paired of colorectal cancer tissues and corresponding adjacent non-cancer tissues through RT-PCR and real time RT-PCR, Results Among 15 testis- enriched genes,found 5 testis-specific expression genes (PRM2,PRM1,TNP1,AKAP4 and TCPll) in 15 human normal tissue genes. Of which PRM1 gene was significantly unregulated in 50% (6/12) of colorectal cancer tissues (more than 2 folds, P(0. 01) as compared with corresponding adjacent non-tumor tissues through real time RT-PCR, Conclusion PRM1 may be a potential CT gene and plays an important role in the molecular diagnosis or immune treatment in colorectal cancer.
