摘  要:目的研究氯通道ClC-3和Cyclin D1在鼻咽癌细胞周期调控中的相互关系,探讨ClC-3参与细胞周期调控的可能机制。方法MTT法检测ClC-3反义寡核苷酸对鼻咽癌细胞增殖能力的影响,流式细胞仪测定细胞周期分布。用反义技术分别抑制细胞内ClC-3和Cyclin D1的表达,用RT-PCR检测目的基因的表达情况。结果40μg/ml ClC-3反义寡核苷酸能明显抑制鼻咽癌细胞的增殖,胞内ClC-3的表达下调对Cyclin D1表达没有影响,而Cyclin D1的表达下调则抑制鼻咽癌细胞ClC-3的表达。结论氯通道ClC-3参与了鼻咽癌细胞的周期调控,Cyclin D1可能位于该调控通路的上游。Objective To study the relation between ClC-3 and cyclin D1 in the cell cycle of human nasopharyngeal carcinoma cell. Methods MTT assay was applied to detect cell proliferation. Cell cycle distribution was analyzed by the flow eytometry. Antisense technology was used to inhibit the expression of endogenous ClC-3 and Cyclin D1. RT-PCR detected the expression levels of destination gene. Results 40μ g/ml ClC-3 antisense oligonucleotides suppressed obviously CNE-2Z cell proliferation.The expression of ClC-3 mRNA was down-regulated in CNE-2Z cells treated with Cyclin D1 antisense oligonucleotides. Conclusion ClC-3 play an important role in CNE-2Z cell cycle. Cyclin D1 may be situated in the upstream of regulation path.
