Effect of Electroacupuncture of Different Acupoints on Plasma and Cerebral Endothelin and CGRP Contents in Acute Cerebral Ischemia Rats

摘  要:目的:探讨督脉取穴法("水沟""百会"穴)和背俞穴取穴法("肝俞""肾俞"穴)抗急性脑缺血损伤的作用机制,并比较两者的针刺效应。方法:将雄性SD大鼠60只随机分为假手术组、模型组、水沟-百会组、肝俞-肾俞组,每组15只。采用颈外动脉插入线栓法建立大鼠大脑中动脉局灶性脑缺血(MCAO)模型,电针参数为疏密波,2/30Hz,电流2mA,持续30min,每12小时1次,共电针6次。假手术组及模型组仅作相应时间点捆扎,不进行针刺。观察神经行为学评分;放射免疫法测血浆和脑组织中血管内皮素(ET)和降钙素基因相关肽(CGRP)含量。结果:大鼠MCAO急性脑缺血后,与假手术组比较:神经功能障碍加重,血及脑组织ET含量明显升高,而CGRP含量明显降低(P<0.05或P<0.01)。大鼠MCAO急性脑缺血后,针刺各组与模型组比较:电针水沟-百会组及肝俞-肾俞组对行为学评分有不同程度的改善,并在缺血48h后产生效应;脑组织ET含量降低(P<0.01),但对血浆ET含量无明显影响(P>0.05);血浆及脑组织CGRP含量升高(P<0.05或P<0.01)。两电针组之间上述结果比较,差异无统计学意义(P>0.05)。结论:(1)电针水沟-百会组和肝俞-肾俞组能有效改善行为学评分,对大鼠MCAO急性脑缺血损伤有一定的保护作用;(2)电针水沟-百会组和肝俞-肾俞组通过降低ET含量和促进CGRP合成、分泌及释放来发挥其对急性脑缺血损伤的保护作用,且两组针刺效应相近。Objective To investigate the underlying mechanism of electroacupuncture (EA) of "Shuigou" (GV 26) and "Baihui" (GV 20), "Ganshu" (BL 18) and "Shenshu" (BL 23) in resisting cerebral ischemia COl). Methods Sixty male SD rats were randomly divided into pseudo-operation group, model group, GV26-GV20 group, and BL18-BL23 group, with 15 cases in each. OI model was established by middle cerebral artery occlusion (MOAO). EA (2/30 Hz,2 mA) was applied to GV26, GV20, BL18 and BL23 for 30 min every time and for 6 times. Neuroethologic scores of the rats were evaluated, plasma and cerebral endothelin (ET) and calcitonin gene-related peptide (OGRP) contents were determined by radioimmunorassay. Results 1 ) After MOAO for 72 hours, the rats' neuroethologic score, plasma and cerebral ET contents in model group increased significantly in comparison with those of pseudo-operation group (P〈0.05, 0.01 ), and plasma and cerebral OGRP levels decreased considerably (P〈0.01). Oompared with model group, the neuroethologic scores, cerebral ET contents of GV26-GV20 and BL18- BL23 groups decreased significantly after OI ( P〈0l01 ) ; and plasma and cerebral OGRP levels in two EA groups increased significantly ( P〈0.05, 0.01 ). No significant differences were found between two EA groups in these 3 indexes ( P〉0.05). Conclusion EA of GV26-GV20 and BL18-BL23 can improve OI rats' behavior activity, which may be related to its effects in lowering cerebral ET and raising blood and cerebral CGRP levels.
