摘 要:目的:研究"松友饮"对高转移人肝癌MHCC97H细胞侵袭性和小鼠免疫功能的影响及其机制。方法:用不同质量浓度的"松友饮"(2、4、8 mg/mL)分别干预MHCC97H细胞48 h,用明胶酶谱实验及侵袭实验研究基质金属蛋白酶-2(matrixmetalloproteinase-2,MMP-2)活性和侵袭性变化。用高转移人肝癌裸鼠原位移植瘤模型进行体内实验,以检测对"松友饮"对肿瘤的抑制作用。C57BL/6小鼠和昆明种小鼠均随机分为"松友饮"剂量递增的C1、C2和C3组及阴性、阳性对照组,灌胃给药,每天1次,连续7 d,处死小鼠后检测自然杀伤细胞(natural killer cell,NK)的活性,观察小鼠腹腔巨噬细胞的吞噬功能变化。结果:与对照组相比,"松友饮"4、8 mg/mL组细胞侵袭性下降有统计学意义(P<0.01),对应的MMP-2活性下降。此外,"松友饮"抑制了肝癌细胞生长,C1、C2和C3组的NK细胞特异性抑制百分率分别为24.6%、27.1%和27.7%,巨噬细胞吞噬百分率与对照组相比,差异有统计学意义(P<0.01)。结论:中药复方"松友饮"可能通过抑制MMP-2活性来抑制肝癌侵袭性,连续7 d"松友饮"灌胃给药能增强小鼠免疫功能。[ ABSTRACT] Objective:To investigate the effects of "Songyou Yin" on the invasiveness of high metastatic human hepatocellular carcinoma (HCC) cell line MHCC97H and immune function of mouse and explore its action mechanism. Methods:The MHCC97H cells were treated with "Songyou Yin" at different concentrations (2, 4, and 8 mg/mL) for 48 h. The Boden chamber assay was ap- plied to determine the tumor cell invasive potential and gelatin zymography was performed to detect activity of matrix metalloproteinase-2 (MMP-2). The nude mouse model bearing HCC xenograft with high metastatic potential was established for the in vivo study. Thirty C57BL/6 mice were randomized into intervention groups of C1, C2 and C3 with stepwise increased dosage of "Songyou Yin", and an- other two groups were regarded as negative and positive controls with 6 mice in each group. Forty Kunming mice were randomized into C1, C2, C3 and control groups with 10 mice in each group. The mice were administered "Songyou Yin" i.g. once daily for7 consecutive days. After the mice were sacrificed the activities of natural killer cells were detected and the phagocytic function of abdominal cavity macrophages was observed. Results.The invasiveness of MHCC97H cells in "Songyou Yin" groups (4 and 8 mg/mL) decreased significantly compared with the control group ( P 〈 0.01 ), which was in accordance with the decreased MMP-2 activity. Moreover, "Songyou Yin" inhibited the growth of HCC in vivo. Cell-specific inhibitory ratio of NK cells was 24.6%, 27.1%, and 27.7% in C1, C2 and C3 groups; whereas phagocytic percentage of macrophages was significantly different from control group (P 〈 0.01 ). Conclusion:Chinese complex herb "Songyou Yin" inhibited the invasiveness of MHCC97H cells by suppressing MMP-2. Intragastric administration of "Sougyou Yin" for 7 consecutive days enhanced the immune function of mice.
