摘  要:目的观察异体角膜体外对人外周血T细胞及其亚群CD28分子表达的调节。方法应用异体角膜与外周血淋巴细胞体外共同培养模型和流式细胞分析技术(fluoresceinactivatedcelsorter,FACS)。结果对照组T细胞CD28表达为21.3%;受角膜或沸波醇酯(phorbol12,13—dibutyrate,PDB)激活后,T细胞CD28表达分别为52.4%和81.4%;受角膜激活后,CD4和CD8T淋巴细胞CD28表达分别为63.1%和52.3%。结论角膜组织体外能够刺激人外周血T细胞CD28表达和T细胞活化,CD4T细胞比CD8T细胞活化更明显。ObjectiveTo observe the immunoregulation of allogeneic cornea to the activation of human peripheral blood T lymphocytes in vitro,and CD 28 expression of T lymphocytes and its subsets when T lymphocytes were stimulated by allogeneic cornea or phorbol 12,13—dibutyrate(PDB).MethodsCulture of human peripheral blood lymphocytes in vitro was studied with allogenic cornea or PDB serving as the stimulative antigen.After co culture for 72 h,T lymphocytes were labelled by monoclonal antibody,and analysed by fluorescein activated cell sorter(FACS).ResultsCD 28 expression on T lymphocytes in control was 21.3%;After stimulation by the allogeneic cornea,CD 28 expression on T lymphocytes was 52.4 %;After stimulated by the PDB,CD 28 expression on T lymphocytes was 81.4%;After stimulated with cornea,CD 28 expression on CD + 4 or CD + 8 subset T lymphocytes was 63.1% or 52.3%.ConclusionAllogeneic cornea can stimulate rather than suppress CD 28 expression on human peripheral blood T lymphocytes in vitro and activation of T lymphocytes.CD 28 can enhance the signal delivery during the activation of T lymphocytes.The activation on CD + 4 T lymphocytes is more prominent than on CD + 8T lymphocytes.
