摘  要:目的测定培养的人眼小梁细胞糖胺多糖(glycosaminoglycan,GAG)的构成比。方法在体外培养从无眼病尸体眼取得的小梁细胞(trabecularmeshworkcel,TMC),掺入氚标葡萄糖胺,再用生物化学方法测定细胞层及培养液中的糖胺多糖,包括依次酶降解、醋酸纤维电泳及液体闪烁放射活性测定等。结果共获得5种糖胺多糖,其构成比是透明质酸30.5%、硫酸软骨素21.7%、硫酸皮质素14.9%、硫酸类肝素19.2%、硫酸角质素5.4%及其他物质8.3%。结论此结果与直接从人眼小梁细胞组织所测得的糖胺多糖的构成比颇为接近,可为进一步研究各种因素对人眼小梁细胞糖胺多糖代谢的影响及开角青光眼的发病学提供基础。ObjectiveTo determine the composition of glycosaminoglycans(GAGs) of cultured trabecular meshwork cells(TMC).MethodsTMC was cultured from fresh cadaver eyes without history of ocular diseases.After incorporation of 3H glycosamine as radioactive precursor,GAGs were extracted from the cell layer and medium with biochemical methods,including sequential degradation by different enzymes,cellulose acetate electrophoresis,and radioactive assay by scintillation counting.ResultsFive GAGs were isolated:hyaluronic acid (HA)30.5%,chondroitin sulfate(CS) 21.7% ,dermatan sulfate (DS)14.9%,heparin sulfate(HS)19.2%,keratan sulfate(KS)5.4% and others 8.3%.ConclusionThe constitution of GAGs synthesized by TMC cultured in vitro is similar to that was found in trabecular meshwork tissues.This report offers a basis for further study of the influence of various factors on GAG synthesis by TMC and the pathogenesis of primary open angle glaucoma.
