摘  要:目的探讨不同病程时期和不同剂量的AT1R拮抗剂对糖尿病视网膜病变(DR)的治疗作用。方法80只龄雄性Wistar大鼠,随机分为DR组(8只)和缬沙坦干预组,干预组按剂量分为大、中、小三组(各24只),每种剂量按给药时间分为第4周开始、第8周开始、第12周开始给药,三组分别给于缬沙坦40mg、10mg及4mg/kg体重灌胃治疗。给药后4周时各组对视网膜中AngⅡ和血管内皮细胞生长因子(VEGF)、组织转化因子β(TGF-β)、胰岛素样生长因子(IGF-1)及血小板衍生生长因子(PDGF)进行平均灰度和密度分析。结果缬沙坦治疗组大鼠同DR对照组比较,前者视网膜中VEGF,TGF-β,IGF-1及PDGF组显著降低,尤其以40mg/(kg.d)组更为明显。结论早期应用大剂量AT1R拮抗剂可一定程度改善DR,对视网膜起到保护作用。Objective To evaluate the effect of valsartan on several growth factors involving into diabetic retinopathy. Methods Eighty male Wistar rats were obtainedm and were randomly divided into diabetic retinopathy (DR, n = 8 ) control group and treatment groups with valsartan 40,10 and 4 mg/(kg · d) by intragastric administration. The treatment were began after 4, 8, 12 weeks. The instrument for computer image - analysis was used to analyze the expression of vascular endothelial growth factor (VEGF), transgorming growth factor- β (TGF- β), insulin -like growth factor ( IGF - 1 ) and platelet derived growth factor (PDGF) in ganglion cell layer of retina. Results After treatment with 4 and 10 mg/kg valsartan, the expressions of VEGF, TGF - β, IGF - 1 and PDGF were reduced than diabetic controls and further reduced with 40 mg/kg of valsartan. Conclusion During the early phase of DR , treatment of angiotensin 1 type receptor ( AT - 1 R) antagonists could exert a potentially therapeutic effect upon the retina of DR.
