托吡酯对戊四氮致癫痫间大鼠海马AQP4表达水平的影响

摘　　要：目的探讨托吡酯对戊四氮致癫痫间大鼠海马AQP4表达水平的影响。方法将30只Wistar大鼠随机分为戊四氮致癫痫间组、托吡酯干预组和正常对照组,每组各10只;癫痫间模型点燃后在不同时相点灌注取材,通过HE染色观察大鼠海马神经元的变化,并应用免疫组化法检测大鼠海马AQP4表达水平。结果HE染色显示托吡酯干预组神经元变性和坏死较戊四氮致癫痫间组明显减轻;免疫组化显示戊四氮致癫痫间组在致癫痫间后12hAQP4的表达显著增强,致癫痫间后24h达高峰,托吡酯干预组在致癫痫间后12h^36h各时相点AQP4表达水平均分别低于戊四氮致癫痫间组相应时间点(P<0.05)。结论托吡酯通过下调大鼠海马AQP4的表达可能参与了对大鼠海马神经元的保护过程。Objective To investigate the effect of topiramate on AQP4 expression of hippocampus in seizure rats induced by pentylenetetrazole （PTZ）. Methods Tirty adult Wistar rats were randomly divided into control （c）group,PTZ group and TPM group after kindling of acute epilepsy , then perfused to acquire the brain at the different point . The hippocampus was studied histologically with HE staining and immunohistochemically for the expression of AQP4. Results Neuronal degeneration and necrosis in CA1 .CA3 and DC areas of the hippocampus were more remarkable histologically in the pentylenetrazole - treated group than that in the topiramate plus pentylenetrazole treated group According to HE method. The expression of AQP4 positive cell in hippocampus in PTZ group increased gradually afer 12 hour, then in the peak afer 24 hour, and compared with TPM group , it was significantly different （P〈0. 05）. Conclusions Topiramate has neuroprotective effect, which might be related with the decreased expression brought by the application of topiramate.
