摘  要:目的探讨血液犬尿氨酸(Kyn)测定时标本采集与保存对测定结果的影响。方法采用高效液相色谱-荧光法测定血清及相应血浆标本中Kyn浓度,对结果进行比较;测定在不同温度下保存不同时间的血清标本中Kyn浓度,对结果进行统计分析。结果EDTA-K2抗凝血浆与血清中犬尿氨酸测定结果比较无统计学意义(P>0.05)。冰冻(-20℃)条件下一周内血清中的Kyn浓度保持稳定。结论在血Kyn测定时,可用EDTA-K2抗凝血浆标本代替血清。血清标本不能及时检测时,最好置于≤-20℃条件下保存。Objective To investigate the effect of sample collection and storage conditions on kynurenine measurements in blood samples. Methods Kynurenine concentration was determined by high- performance liquid chromatography - determination of fluorescence (HPLC- FLD) using anticoagulant EDTA - K2 plasma or serum. Preserved at differen1 temperatures for varied time, serum specimens were analyzed by HPLC - FLD and the determination results were compared. Results There was no statistically significant difference between EDTA- K2 anticoagulant plasma and serum (P 〉 0.05 ), and Kyn concentration remained stable under the condition of 20℃ within one week in the serum. Conclusion It is feasible to replace serum with EDTA - K2 anticoagulant plasma. Serum sample should be kept under the condition of ≤ - 20℃within one week.
