摘  要:背景:移植后造血干细胞的顺利归巢是单倍相合骨髓移植成功的关键。目的:构建小鼠白血病模型,进行单倍型相合的骨髓移植,动态观察受体小鼠体内移植细胞归巢率及不同时间点的归巢规律。设计、时间及地点:随机对照观察实验,于2005-11/2007-10在福建医科大学实验动物中心清洁级动物实验室进行。材料:供鼠为清洁级近交系C57BL/6小鼠和C57BL/6小鼠与BALB/c小鼠杂交的雄性F1小鼠,受鼠为清洁级近交系雌性BALB/c小鼠,用于构建白血病小鼠模型。方法:将48只造模小鼠随机分为2组,对照组供鼠为C57BL/6小鼠,实验组供鼠为F1小鼠。实验组进行主要组织相容性复合物单倍型相合的同种异基因骨髓移植,对照组进行主要组织相容性复合物完全不相合的骨髓移植。主要观察指标:移植后1,2,3,4d动态观察小鼠外周血、骨髓和脾中有核细胞计数,以流式细胞仪检测可与FITC标记的抗鼠H-2Kb单抗结合的供鼠阳性细胞并计算骨髓和脾归巢率。结果:对照组外周血中供鼠来源的细胞数持续下降,脾和骨髓的归巢率均为先升后降,提示供者来源的骨髓细胞在受鼠体内为先归巢再出巢;实验组外周血中供鼠来源的细胞数先下降,再持续上升,骨髓归巢率为下降后上升,脾归巢率为上升后下降,提示供鼠来源骨髓细胞在骨髓中表现为出巢后再归巢,脾中有延迟,与对照组比较,实验组第4天外周血中供鼠细胞数量及骨髓和脾归巢率均增高(P<0.05)。结论:主要组织相容性复合物单倍体相合的骨髓移植的归巢规律是先出巢再归巢,归巢的细胞数量和归巢率高于主要组织相容性复合物完全不相合骨髓移植。BACKGROUND: Successful homing of haemopoietic stem cells is the key step for the success of haploidentical bone marrow transplantation. OBJECTIVE: A mouse model of leukemia was constructed, and then the haploidentical bone marrow transplantation was performed and the homing ratio and characters of homing at different time points were investigated dynamically in the recipient mice. DESIGN, TIME AND SETTING: A randomized control experiment was performed at the Cleaning Animal Lab of the Animal Experimental Center in Fujian Medical University from November 2005 to October 2007. MATERIALS: The donor mice were the male F1 hybridism mice of cleaning inbred strain C57BL/6 and BALB/c mice or the cleaning C57BL/6 mice. The recipient was the cleaning female inbred strain BALB/c mice. These mice were used for establishing leukemia models. METHODS: Totally 48 mice were divided into 2 groups. The donor was C57BL/6 mice in the control group and F1 hybridism mice in the experimental group. MHC haploidentical allogeneic bone marrow transplantation was performed in the experimental group and MHC mismatched bone marrow transplantation in the control group. MAIN OUTCOME MEASURES: Nucleated cell number in peripheral blood, bone marrow and spleen was counted; the percentage of positive cells capable of connected with FITC labeled anti-mouse H-2Kb antibody was detected by flow cytometry and the homing ratio in bone marrow and spleen was calculated dynamically at days 1, 2, 3 and 4 after transplantation. RESULTS: In the control group, the amount of myeloid cells in peripheral blood kept descending and the homing ratio in bone marrow and spleen increased then descended, which showed that the myeloid cells of donor presented homing then mobilizing in the body of recipient. In the experimental group, the amount of myeloid cells in peripheral blood firstly descended, then kept increasing; the homing ratio descended then increased in bone marrow and increased then descended in the spleen, which indicated that the m
