摘 要:目的:观察大鼠脊髓缺血再灌注损伤(SCI)过程中脑红蛋白(Ngb)的表达变化及其意义。方法:健康成年Wistar大鼠随机分为脊髓压迫缺血再灌注组和假手术对照组。运用原位杂交组织化学和免疫印迹法,检测压迫段脊髓组织中Ngb的表达变化。结果:再灌注2h后,Ngb在压迫段脊髓开始表达上调,24h较为明显,48h达峰值,72h表达减少,与假手术组相比较,具有显著性差异。免疫印迹法的结果和上述结果一致。结论:在脊髓缺血再灌注损伤中Ngb的表达变化与再灌注时间相关,其表达上调是机体内源性神经保护之一。Objective : To evaluate the changes of neuroglobin (Ngb) expression after spinal cord ischemia/reperfusion injury (SCI) in rats. Methods: Fifty-six adult Wistar rats were randomly divided into 6 groups, including sham operation group (n=6) and five operation groups (n= 10). The spinal cord SCI model was established with a self-made device in the operation groups, The expressions of Ngb mRNA and protein levels were detected by using in situ hybridization histochemistry and Western blotting in the spinal cord tissue respectively. Results : The expression of Ngb mRNA was increased at 2 h after SCI, and peaked at 48 h, but appeared to reduce significantly after reperfusion for 72 h. The Ngb mRNA optical densities at 2, 12, 24, 48 h and 72 h groups were (0. 26±0. 05), (0. 28±0. 04), (0.30±0.05), (0. 41±0. 08) and (0. 32±0. 06) respectively and significantly higher than those in the control group (P〈0.05). Conclusion : The change of Ngb following SCI in the rats presents a time-dependent manner, which may contribute to the endogenous protection.
