摘  要:目的:探讨胰岛素样生长因子-1(IGF-1)对雪旺细胞氧化损伤的保护作用。方法:用体外纯化培养的雪旺细胞建立氧化损伤模型,将培养细胞分成氧化损伤组、IGF-1保护组和正常对照组。四甲基偶氮唑蓝比色法(MTT)检测细胞的活性,生化技术检测细胞内超氧化物歧化酶(SOD)的含量,免疫印迹法检测凋亡蛋白Bcl-2的表达水平。结果:与对照组相比,H2O2处理组细胞胞体积缩小、空泡化,细胞活性降低,SOD含量明显减少,Bcl-2表达减弱;而IGF-1保护组细胞存活率明显升高,SOD含量较损伤组高,Bcl-2表达明显上调。结论:IGF-1对氧化损伤的雪旺细胞有保护作用。Objective: To investigate the protective effects of the insulin-like growth factor-1 on the oxidative injured Schwann cells. Methods: Cultured Schwann cells were randomly divided into experimental groups and control groups, the former were subdivided into hydroqen peroxide treated group and insulin-like growth factor-1 preconditioned group. MTT was used to observe the viability of the Schwann cells. The content of intracellular superoxide dismutase (SOD) was detected by a biologic chemistry method. Westem blotting was used to detect the expression of Bcb2. Results: Compared with the control group, the survival Schwann cells decreased in the oxidative injured group. However, these changes were significantly inversed in the insulin-like growth factor-1 preconditioned group. Conclusion: The insulin-like growth factor-1 has a good antioxidant effect on the Schwann cells.
