IP_6对人结肠癌细胞HT-29凋亡Caspase3基因作用

摘　　要：目的研究肌醇六磷酸（IP6）对人结肠癌细胞HT-29的诱导凋亡作用,检测Caspase3基因在IP6处理细胞前后表达的变化,并对IP6诱导人结肠癌细胞HT-29的凋亡机制进行初步探讨。方法不同浓度的IP6以不同时间作用于体外培养的HT-29细胞,用MTT法检测IP6持续作用对癌细胞增殖的影响;透射电镜观察癌细胞超微结构的变化;用RT-PCR方法对IP6作用前后以及作用不同浓度和时间的癌细胞内Caspase3活性进行检测。结果IP6能显著抑制人结肠癌细胞HT-29生长,呈剂量与时间依赖性;IP6作用后,HT-29细胞出现凋亡的形态学改变;与对照组相比,不同浓度的IP6作用不同时间后,Caspase3 mRNA表达均升高（F=8.173-91.755,q=3.960-17.095,P〈0.05）,IP6作用6个月组Caspase3 mRNA表达升高较其他时间组显著（q=12.858-14.890,P〈0.01）。结论IP6具有抑制人结肠癌细胞HT-29增殖和诱导凋亡的作用;IP6长期作用诱导细胞凋亡更加显著;Caspase3基因参与细胞的凋亡调节机制,发挥重要作用。Objective To study the mechanisms of apoptosis induced by Inositol Hexaphosphate （IP6） in HT-29 human colon carcinoma cell line. Methods HT-29 cells cultured in vitro were exposed to various concentrations of IP6 applied at different time. The constant effect of IP6 on HT-29 ceils proliferation was evaluated by MTT assay. The morphological changes of HT- 29 cells were observed under electronic microscope. The activity of Caspase3 in HT-29 cells was detected by RT-PCR assay. Results IP6 remarkbly suppressed the growth of HT-29 cells. The suppression was a dose-dependent and time-dependent manner. HT-29 cells showed morphological changes of apoptosis after IP6 treatment. Compared with the control group, the expression of Caspase3 mRNA was significantly increased at all IP6 concentrations （F= 8. 173-91. 755 ,q= 3. 960-17. 095, P〈0. 05）. The upregulation of Caspase3 mRNA expression was significantly higher in the 6-month group than the rest groups （q= 12. 858-14. 890, P〈0. 01）. Conclusion IP6 can suppress the proliferation and induce the apoptosis of HT-29 cells. Longer IP6 treatment induced stronger apoptosis. Caspase3 plays an important role in the regulatory mechanisms of cells apoptosis.
