术前短期应用血管生长抑制剂人参皂甙Rg3对膀胱肿瘤组织微血管密度变化的影响

摘  要:目的观察术前短期应用血管生长抑制剂人参皂甙Rg3对膀胱肿瘤组织微血管密度(MVD)变化的影响。方法选取30例膀胱肿瘤患者,结合CT和膀胱镜检查活检的病理资料明确诊断,留取膀胱肿瘤组织标本。患者术前给予血管生长抑制剂人参皂甙Rg3,20 mg/次,每日2次,口服,连续用药10~14天。10~14天后手术,术后留取肿瘤组织标本。肿瘤组织切片经免疫组化染色,标记抗体。利用Le ica Qw in V3图像分析系统对染色切片进行分析,计算肿瘤组织MVD,比较用药前后肿瘤组织MVD的变化。结果用药后肿瘤组织MVD明显低于用药前〔(1.49±1.26)×10-4/μm2vs.(4.43±2.26)×10-4/μm2,P〈0.05〕。结论术前短期应用血管生长抑制剂人参皂甙Rg3能明显降低膀胱肿瘤组织MVD。Objective To observe the effects of short - term preoperative administration of angiogenesis inhibitor ginsenoside Rg3 on the variation of microvessel density (MVD) of urinary bladder tumor. Methods Specimens of urinary bladder tumors were obtained by biopsy from 30 patients diagnosed with urinary bladder tumor, based on combined examinations of CT and cystoscopic biopsy. The 30 patients were each given a dose of 20 mg of ginsenoside Rg3 orally twice daily for 10 to 14 days preoperatively. Postoperatively, specimens of tumor tissues were retained for sections, followed by immunohistochemistry staining and antibody conjugates. The sections were analyzed by an image analysis system (Leica Qwin V3 ) to calculate the MVD of tumors and to compare the variations of the MVD of tumors before and after medication. Results The MVD of the tumor tissues after the medication was markedly lower than before [ ( 1.49 ± 1.26 ) × 10^-4/μm^2 vs. (4.43±2.26 )×10^-4/μm^2 ,P 〈 0.05 ). Conclusion Short - term preoperative administration of anglogenesis inhibitor ginsenoside Rg3 can markedly reduce the MVD of tumor in patients with urinary bladder tumor.
