摘  要:目的探讨利福平对鱼藤酮所诱导的人多巴胺能神经母细胞瘤细胞SH-SY5Y凋亡的抑制作用。方法采用100nmol/L鱼藤酮作用SH-SY5Y细胞24h,建立SH-SY5Y细胞凋亡模型;再用100、200、300μmol/L不同浓度的利福平进行干预,采用MTT比色法检测细胞活性,采用流式细胞术(FCM)及原位缺口末端标记染色法(TUNEL)检测细胞凋亡,Western blot法检测前凋亡蛋白3-磷酸甘油醛脱氢酶(GAPDH)的表达水平。结果用100nmol/L鱼藤酮作用SH-SY5Y细胞24h,能够诱导该细胞产生凋亡并降低其活性;利福平干预后,鱼藤酮所诱导的SH-SY5Y细胞凋亡被抑制,并且这一抑制作用与利福平浓度有明显的剂量-效应关系;而且利福平干预后能降低GAPDH蛋白的表达水平。结论利福平对鱼藤酮诱导的SH-SY5Y细胞凋亡具有明显的抑制作用,这一抑制作用可能是通过下调GAPDH蛋白的表达而得以实现。Objective To study the inhibitory effect of rifampicin on rotenon-induced apoptosis of SH-SY5Y cells. Methods SH-SYSY cells were incubated with 100 nmol/L rotenon for 24 h to induce cell apoptosis. Rifampicin with different concentrations (100, 200 and 300 umol/L) was added respectively to each experimental group. MTT assay was used to measure cell viability, flow cytometry and TUNEL assay were used to test cell apoptosis, and Western blot was used to detect the expression of GAPDH. Results Rotenon could induce apoptosis of SH-SYSY cells and decrease cell viability. After treatment with rifampicin, apoptosis of SH-SYSY cells induced by rotenon was inhibited in a concentration-dependent manner and the expression of GAPDH protein was reduced. Conclusion Rifampicin can inhibit the apoptosis induced by rotenon in SH-SYSY cells by down- regulating the expression of GAPDH protein.
