摘 要:[目的]探讨建立肾癌ACHN细胞系裸鼠皮下移植瘤及转移瘤模型的适宜条件。[方法]MTT法观察细胞生长情况。将裸鼠分组,分别用2.0×106/100μl(A组)、3.0×106/100μl(B组)和4.0×106/100μl(C组)的细胞接种浓度,注入裸鼠后项中间皮下;用4.0×106/100μl的细胞接种浓度,分别接种于裸鼠后项中间(C组)、背部(D组)和前肢腋窝皮下(E组);分别用第20代(F组),第30代(G组)和第10代(C组)的肿瘤细胞4.0×106/100μl,注入裸鼠后项中间,观察各组成瘤情况。尾静脉注射瘤细胞1.0×106/ml建立转移模型。[结果]第4天细胞进入对数生长期。A组成瘤率为33.3%、B组和C组的成瘤率均为100%,但C组的成瘤速度明显高于B组;C组、D组和E组的成瘤率均为100%,注射肿瘤细胞后第10天,C组、D组和E组的肿瘤生长平均相对体积,各组间尚无明显差异;注射后第30天,C组的肿瘤生长平均相对体积显著高于D组和E组(P<0.05);G组的成瘤率为83.3%,F组的成瘤率为100%,与C组相比,无显著性差异(P>0.05)。尾静脉注射组全部出现转移灶。[结论]使用ACHN细胞株,控制好浓度、接种部位以及传代次数后能获得较理想的裸鼠肾癌皮下移植瘤模型;使用传统尾静脉注射法可以建立裸鼠转移模型。[Purpose] To investigate the suitable condition of establishing subcutaneously transplanted and metastatic nude mice model with human renal carcinoma of ACHN cells. [Methods] MTT method was used to detect the proliferation of ACHN cells. The cells with concentration of 2.0×10^6/100μl(group A), 3.0×10^6/100μl(group B) and 4.0×10^6/100μl(group C) were inoculated respectively into collare of nude mites subcutaneously. ACHN cell with concentration of 4.0×10^6/100μl was injected into nude mice's collate (group C), back (group D) and subcutaneous layer of forelimb axillary fossa(group E) respectively. The 20th generation(group F), the 30th generation (group G) and the 10tb generation(group C) tumor cells with 4.0×10^6/100μl were inoculated into collate of nude mice subcutaneouly respectively. The growth of tumor was examined in all groups. The ACHN cell concentration with 1.0×10^6/ml was injected into nude mice through vena caudalis to establish cancer model. [Results] After 4 days, cell entered the logarithmic growth phase. The rate of tumor formation was 33.3% in group A, 100% in groups B and C, while the speed of tumor growth significantly increased in group C eompared with group B. the rate of tumor formation was 100% in groups C, D and E. After 10 days, mean relative tumor volume among the three groups (C, D, E) were no significantly different. But 30 days after injection, tumor mean relative volume of group D and E were significantly less than that of group C; the rate of tumor formation was 100% in group F, 83.3% in group G, with no significant difference compared with group C (P〉 0.05). Metastatic tumor model was established successfully in all nude mice. [Conelusionl Using ACHN cells with suitable concentration, proper injective site, can successfully create subcutaneously transplanted renal cell earcinoma model in nude mice after several subcultures. Nude mice metastatic tumor model can be established through traditional vena caudalis injection.
