Glut1、VEGF、HIF-1α在乳腺癌中的表达和临床意义

摘　　要：目的探讨Glut1在乳腺癌中的表达及与VEGF和HIF-1α表达的关系和临床意义。方法建立负乳腺癌(MCF-7)裸鼠模型,不同大小乳腺癌病灶,行免疫组化S-P法,分别检测Glut1、VEGF和HIF-1α的表达,分析三者表达的相关性。并且与同部位正常组织的Glut1、VEGF和HIF-1α免疫组织化学结果进行比较。结果Glut1、VEGF、HIF-1α在乳腺癌中表达的阳性率分别为43.3%、50.0%和33.3%,明显高于正常组织(P<0.05)。一定范围内,三者的表达与肿瘤生长的大小有关(P<0.02)。Glut1、VEGF和HIF-1α三者表达有一致性,VEGF和HIF-1α的表达都与Glut1呈正相关(r分别为r=0.503,r=0.668)。结论Glut1是一项综合指标,可直接反映肿瘤的代谢水平,可间接反映肿瘤转移可能性大小及乏氧程度,对临床肿瘤的诊断、分期和预后有重要的指导意义。[Objective] To study the expression of Glut1, VEGF, HIF-1α in breast carcinoma and their relation and significance in clinic. [Method] The naked rat model bearing breast earcinoma（MCF-7） was established, and expressions of Glut1, VEGF, HIF-1α in different size breast carcinoma tissues and normal tissues were detected and analyzed by immunohistochemistry S-P method. [Result] The positive rates of Glut1, VEGF and HIF-1α protein in the tumor were 43.3%, 50.0%, 33.3%, respectively. And compared to normal tissues, there was a significant difference （P 〈0.05）. There exist the unity and correhtion（r =0.503, r =0.668） among the expressions of Glut1, VEGF, HIF-1α and their variation with the size of tumor （P 〈0.05） in a scope. [Conclusion] Glut1 not only reflects the glucose metabolism but also reflects the possibility of tumor invasion and the degree of hypoxia. So it can act as a fundamental guide to clinical diagnosis, staging and prognosis of the tumor.
