Establishment of a complete cleft palate animal model by surgical technique

摘  要:目的选择合适的外科手术方式,建立长期、稳定的完全性腭裂骨缺损动物模型。方法取2个月龄山羊10只为实验对象,随机分成两个实验组,以两种不同的外科手术技术,形成硬腭部0.5cm×10cm全层、全长硬腭骨裂隙。分别在术后1周和32周,观察裂隙软、硬组织形态,并拍摄头颅CT及三维重建。结果两个实验组山羊手术后均成活,健康状况良好,术后32周后,干骨标本见硬腭裂隙全长仍然存在,无自发骨性愈合,形态上可见裂隙前后较窄中间稍宽,3D-CT测量显示,骨缺损宽度比手术时增大。结论此两种手术方式建立的腭裂骨缺损动物模型,经长期观察,裂隙均不能自行修复,效果相似。建立腭裂动物模型,具有充分的可行性。Objective To establish a complete cleft palate model by selection of suitable surgical approaches. Methods Ten healthy goats at age 2 months were divided randomly into two experimental groups. All goats were operated on and two types of palate osteoectomy were performed. All animals were sacrificed at 32 weeks after operation. The morphology of soft, sclerous tissues of the cleft was measured and compared at 1 and 32 weeks postoperatively by 3D- CT. The state of the cleft in these two groups was also observed directly after cleaning the skulls. Results All goats were in good condition during the experiment. No spontaneous bone healing occurred in experimental animals. The width of the cleft palate at 32 weeks postoperatively was greater than that at 1 week postoperatively in each group, and there was no significant difference between two groups. Conclusion It is feasible to an establish experimental complete cleft palate on young goats by the two types of surgical operations for further study.
