摘 要:目的:观察荜拔油不皂化物对实验性C57BL/6小鼠胆囊结石形成的抑制作用。方法:C57BL/6鼠30只,每组10只,模型周期4周,对照组喂饲普通饮食;模型组喂含1%的胆固醇饮食;用药组喂含1%的胆固醇饮食和荜拔油不皂化物(30mg/kg体重)灌胃。结果:模型组9只出现胆囊结石,10只出现胆固醇结晶;荜拔油不皂化物组1只出现胆囊结石和结晶;对照组无结石和结晶形成。与模型组相比,荜拔油不皂化物组和对照组的肝氨肽酶N(APN)mRNA及胆囊胆汁胆固醇饱和指数(CSI)明显降低。结论:荜拔油不皂化物明显抑制高胆固醇饮食诱发的小鼠胆囊结石的形成,可能通过降低肝脏APN mRNA水平及胆囊胆汁胆固醇饱和指数发挥作用。Objective To explore the effect of the oil of piper longum unsaponifiablc matter(OPUM) on experimental gallstone formation in the gallbladder of C57BL/6 mice. Methods 3 dietary group of C57BL/6 with 10 each were allocated as control (normal mice chow), lithogenic ( 1% cholesterol diet) and OPUM ( 1% cholesterol diet + 30 mg/kg body weight) group respectively for 4 weeks. Results Cholesterol crystals and stones were found in 10/10 and 9/10 gallbladder respectivcly in the lithogcnic group, 1/10 gallstone and 1/10 gallstonc crystal respectively in the OPUM group, but none in thc control group. Comparing with the lithogenic group, the expression of aminopeptidase N (APN) gene in liver tissue and cholesterol saturation index(CSI) in thc gallbladder bile were significantly decreased in OPUM and control groups. Conclusion OPUM inhibits the experimental gallstone formation induced by high cholesterol feeding in C57BL/6 mice, with simultaneous decreasing of APN gene expression in thc liver tissue and of the CSI values in the gallbladder bile may be the cause.
