正常人视网膜抗氧化能力的分析

摘  要:目的:测定正常人视网膜不同部位SOD、GST、MDA水平,探讨视网膜不同部位抗氧化能力的差异性,为视网膜相关疾病的发病机理及防治提供实验及理论依据。方法:取成人眼球,6人(12只眼),生理盐水冲洗,赤道部切开眼球,弃去眼前节及玻璃体,取黄斑部、后极部、周边部全层视网膜三组,制成10%组织匀浆作生物化学检验。分别应用黄嘌呤氧化酶法测定SOD,化学比色法测定GST,硫代巴比妥酸(TBA)法测定MDA。结果:黄斑部、后极部、周边部视网膜SOD、GST含量依次升高,黄斑部最低,比较差异具有统计学意义(p<0.05);MDA含量依次降低,黄斑部最高,比较差异具有统计学意义(p<0.05)。结论:视网膜黄斑部、后极部、周边部三个部位的氧化指标即SOD、GST、MDA存在差异,提示视网膜黄斑部抗氧化能力最弱。Objectlve:To estimate the antioxidant properties in different parts of retina through determining the levels of SOD, GST, MDA, further to investigate pathological mechanism and provide the possible avenue to treat the ophthalmological diseases coming from oxidative stress. Methods: The eyes of 6 humans (who were subjected to suddenly dead) were removed and washed with sterilized saline. Incisions were made at the equatorial plane of eye- balls, and the parts of full retina, including yellow spot ( Y), posterior part (PO) and peripheral part (PE) were removed and homogenized for biochemical analysis, respectively. The levels of SOD was measured with xanthine oxidase method, GST was measured with colorimetric method, MDA was measured with TBA. Results:The levels of SOD and GST in yellow spot, posterior part and peripheral part of retina were revealed in the pattern of Y 〈 PO 〈 PE ( p 〈 0.05 ), while the levels of MDA showed the pattern of Y 〉 PO 〉 PE ( p 〈 0.05 ). Conclusion : Yellow spot, posterior part and peripheral part of retina have different antioxidant properties, and the lest oxidative stress in yellow spot of retina.
