摘  要:本试验采用水煮醇沉淀法研究了影响茶叶多糖(TPS)提取的因素,并利用苯酚-硫酸法测定多糖含量。通过控制和改变提取时间、提取温度、乙醇沉淀体积倍数研究这些因素对提取结果的影响,并比较了Sevag法与三氯乙酸(TCA)法除蛋白质的效果,再利用凝胶柱层析法对TPS进行初步纯化。结果表明,TPS最佳提取的条件为:100℃、4h、2.5倍体积乙醇沉淀效果最佳。粗糖提取率为9.1624%,相对含量为44.6%。TCA法除蛋白质效果较好,经凝胶柱层析,确定本方法所提取的茶叶多糖为一种糖蛋白。In this experiment,the factor on extraction of soluble polysaccharide from tae(TPS)with the method of water dissolving following by ethanol deposit was researched by controlling the factor of extracting time, temperatare, depositing volume times of ethanol.The different reclaiming efficiency was researched by detecting the polysaccharide with the method of vitriol-phenol.At the same time,the purified method of TPS by the method of Sevag and TCA was compared. Then the primary purified TPS was purifid by Sephadex G-75 for further step.The result showed that the best extraction conditions of TPS were that the temprature was 100 ℃,the extraction time was 4 h and the best depositing volume multi- ple of ethanol was 2.5 times.The crude efficiency of extraction was 9.1624 % ,the relative content was 44.6 %.The method of TCA was better than Sevag.By the purifying method for further step of Sephadex G-75 colume chromatography,the production was a sort of glycoprotein.
