复合甲氨蝶呤硫酸钙人工骨缓释剂的制备及其对骨肉瘤细胞的体外抑制作用

摘　　要：目的探讨复合甲氨蝶呤(MTX)硫酸钙人工骨缓释剂的制备方法及对骨肉瘤的治疗作用。方法以硫酸钙人工骨为载体,制备载药量为20 mg、质量百分比为4%的药粒。以Tris缓冲生理盐水作为模拟体液收集20 d内的液体标本,检测其缓释特性。四甲基偶氮唑盐法(MTT)检测20 d内缓释剂浸出液对骨肉瘤细胞MG-63的抑制作用,用酶标仪测吸光度值(A)计算抑制率。结果缓释剂前2 d出现快速释放,此后在较低水平维持相对稳定的缓慢释放,持续20 d以上。其第2、10、18、20天浸出液的MTX水平分别为161.598、97.752、73.274、16.946g/L。对骨肉瘤细胞MG-63的抑制率分别为64.2%、21.0%、13.4%、12.4%。结论硫酸钙人工骨复合MTX具有良好的缓释功能,在20 d内对骨肉瘤细胞MG-63有良好的抑制作用。Objective To develop a drug of Sustained-releasing methotrexate combined with calcium sulfate and to investigate it＇s inhibitory ability on human osteosarcoma cell MG-63. Methods The drug particles combined with 20 mg methotrexate were obtained, 4% in weight percentage, as the calcium sulfate for the carrier. The drug concentration of each sample was analyzed through high performance liquid chromatogram and were extracted in 20 days respectively. The inhibitory action of extracted solution to the human osteosarcoma cell MG-63 was detected by MTT assay. Results Methotrexate was released rapidly during the first 2 days. Then,the drug kept slow and stable release rate and could exist more than 20 days. The methotrexate content of the 2,10,18,20 day was 161. 598,97. 752,73. 274,16. 946 respectively, and it＇s inhibitory ratio on osteosarcoma MG-63 was 64.2%,21.0%,13.4%,12.4% respectively. Conclusions The MTX particle combined with calcium sulfate has good sustained - releasing effect, and which has medium inhibitory effect on osteosarcoma cell MG-63 in 20 days.
