摘　　要：目的研究VCPmRNA表达与胃癌及淋巴结转移的关系,探讨其术前预测淋巴结转移危险度及预后的意义。方法采用RT-PCR、聚丙烯酰胺凝胶电泳EB染色法,检测42例胃癌组织、癌旁正常胃组织、淋巴结组织中VCPmRNA的表达。结果VCPmRNA在上述组织细胞中均有表达,胃癌组织和淋巴结组织明显高于癌旁正常胃组织(P<0.001),胃癌组织VCPmRNA表达丰度随转移淋巴结枚数的增多而增高,转移淋巴结数量≥16枚者高于<16枚者(P<0.05)。结论胃癌组织VCPmRNA过度表达的患者淋巴结转移范围及程度相对较重,癌细胞侵袭力较强,宜行扩大根治术;术前RT-PCR检测胃癌组织VCPmRNA表达有助于确定治疗方案及判断预后。Objective To investigate the relationship between VCPmRNA expression and gastric cancer and lymph node metastasis and to assess the significance of predicting the extent of lymph node metastasis and prognosis. Methods VCPmRNA expression was examined in matched primary lesions, normal gastric mucosa and lymph node metastatic lesions among 42 patients with gastric cancer by reverse transcription polymerase chain reaction（RT-PCR） ,polyacrylamide gel electrophoresis（PAGE）and EB stain with the relation between VCPmRNA expression and clinicopathologic factors analyzed. Results The primary tumor samples showed a significantly higher level of VCPmRNA expression compared with matched normal tissue, so did the lymph node metastatic samples（P〈0. 001）. The VCPmRNA expression showed a low level in benign lesions. The VCPmRNA expression showed a higher level in patients with the number of lymph node metastasis above 16 than below 16（P〈0.05）. Conclusion The degree of lymph node metastasis and invasiveness of carcinoma cells are more serious in cases with VCPmRNA overexpression. The extensive lymph node dissection should be carried out for these patients. Examination of VCPmRNA expression by RT-PCR before surgery is indicated to arrive at an optimum treatment scheme and to estimate the prognosis.
