摘  要:在以往研究苹果锈果类病毒(ASSVd)的基础上,以斑点杂交检测ASSVd呈阳性的杏树为试验材料,利用RT-PCR技术扩增出苹果锈果类病毒全长特异片段,通过回收克隆测序,发现杏树苹果锈果类病毒基因组长为330nt,将克隆测序结果在GenBank中登录,获得10条序列,登录号为EU031487-EU031496。在此基础上,利用原位RT-PCR检测技术,进一步证明杏树叶片中有苹果锈果类病毒存在,主要分布在细胞核中。本研究证实苹果锈果类病毒也能侵染杏树。Present study was conducted based on the previous research of Apple scar skin viroid(ASSVd)for apple and pear,used material of apricot was those that showed positive signal of blot hybridization detection.Full-length special fragment of ASSVd for apricot was amplicated using RT-PCR protocol,then was recycled,cloned and sequenced.It was demonstrated that the full-length genome of ASSVd of apricot is 330 nt long(GenBank numbered with EU031487 to EU031496).The exist of ASSVd in apricots was reconfirmed by the results of in situ RT-PCR,and ASSVd mainly located in nucleolus of mesophyll cell.The fact of infecting capability of apricot enlarged the host ranges of ASSVd.
