摘  要:目的观察大鼠晶状体摘出术后晶状体上皮细胞(lens epithelial cells,LECs)的形态、分布改变及其转分化的动态过程。方法对50只SD大鼠左眼行晶状体囊外摘出术(extracapsular lens extraction,ECLE)。分别于术后0h、3d、7d、14d及28d对术眼进行裂隙灯检查并处死动物各10只,摘除眼球行光镜及免疫组化检测,Western Blot法检测各时间点后囊膜仅.平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)表达。结果术后0h于前囊下和赤道部的内表面观察到LECs,后囊膜无细胞分布。晶状体后囊膜混浊在术后3d出现,囊膜皱缩,整个晶状体囊袋内出现纺锤形细胞。7d时后囊膜明显混浊,囊袋内见较多纺锤形细胞分布。术后14d时出现明显后囊膜皱缩,见新生晶状体纤维,纺锤形细胞明显减少,后囊膜仍可见细胞。28d可见明显后囊膜增厚,新生晶状体纤维填充囊袋,后囊膜未见细胞。免疫组化检测见术后3d α-SMA阳性表达。Western Blot检测α-SMA于术后0h几乎检测不到,3d表达明显增强,7d达高峰,14d下降,28d明显下降。结论大鼠ECLE术后囊袋内LECs形态及分布呈动态变化,后囊膜α-SMA表达提示ECLE术后3~7d是LECs转分化的关键时期。Objective To observe the shape and distribution of lens epithelial cells (LECs) and the dynamic process of their transdifferentiation during the development of posterior capsule opaciflcation (PCO) in rats. Methods Extracapsular lens extraction (ECLE) was performed in 50 rats' left eyes. Animals were observed by slit lamp at 0 hour,3 days,7 days,14 days and 28 days after the surgery,and then 10 rats were killed at each time point. The eyeballs were enucleated for light microscopy observation and immunohistochemistry measurement. The expression of the posterior capsule alpha-smooth muscle actin(a-SMA) was detected by the Western Blot method at each time point. Resuits The LECs were observed in the inner surface of the anterior capsule and lens bow at 0 hour after the surgery,and no cells were observed for the posterior capsule.Three days after the surgery,PCO began to occur and capsular wrinkling was apparent. Some spindle shaped cells were present throughout the lens capsule. Seven days after the surgery, PCO became obvious and a lot of flbrocyte could be seen in the posterior capsule. Posterior capsular wrinkling became obvious at fourteen days after surgery. Regenerated lens fiber could be observed under the light microscope and the number of spindled shaped cells decreased significantly. But cells could still be seen at the posterior capsule. Twenty-eight days after the surgery, the posterior capsule became obviously thicker and the regenerated lens fiber filled the capsular bag. NO cells was found in the posterior capsule. The transdifferentiation of the LECs were indicated by the detection of α-SMA through immunohistochemistry measurement 3 days after ECLE. The Western Blot method indicated that little α-SMA could be detected at 0 hour after the surgery. Three days after the surgery α-SMA increased in the posterior capsule. Seven days after the surgery, it reached the peak and decreased 14 days after the surgery. Twenty-eight days after the surgery, α-SMA decreased dramatically. Conclus
