摘  要:为研究不同类型柿果实的脱涩与乙醇脱氢酶(ADH)之间的关系,采用聚丙烯酰胺凝胶电泳法,对4种类型的5个柿品种不同部位和果实不同发育时期的ADH进行了同工酶分析。结果表明:成熟期的果实中,不同脱涩类型柿果实的ADH均表现2条谱带,完全甜柿和不完全甜柿果实的ADH酶带谱型一致,不完全涩柿果实的ADH酶带中独有一条更靠近负极的谱带,而完全涩柿中则没有检测到ADH同工酶带;果实发育过程中,2个完全甜柿出现酶带的时期较早,分别在7月中旬和8月中旬,不完全甜柿和不完全涩柿则在果实发育后期(8月底和9月中旬)出现酶带,而完全涩柿果实从始至终都没有出现ADH同工酶带。不同器官的ADH同工酶电泳结果表明,果实成熟时在不同器官中共检测到5条ADH同工酶谱带,其中酶带b、酶带d、酶带b和c分别是柿果实、叶片、枝皮的特征性酶带,说明ADH同工酶具有器官特异性。本试验初步研究了柿果实发育过程中ADH在不同器官的分布特点,为进一步研究ADH在柿不同脱涩类型果实脱涩机制中的作用及其相关基因表达提供参考。The patterns of alcohol dehydrogenase isozyme in fruits at various periods and in different organs among four different types of persimmon (Diospyros kaki Thunb) were investigated by polyacrylamide gel electrophoresis (PAGE). The same ADH isozyme patterns were shown in ripe fruits of the pollination constant non-astringent and pollination variant non-astringent type, but the pollination variant astringent type had a distinct ADH isozyme band near the negative pole. The ADH isozyme bands were not detected in the pollination constant astringent type. The ADH isozyme patterns appeared in the early stage (about mid-July to mid-August) of fruit development in pollination constant non-astringent type and in the late stage (about late-August to mid-September) among pollination variant non-astringent and pollina- tion variant astringent type. There were no detectabte ADH isozyme band in the potlination constant non-astringent type. As to the organs, band b, band d, and bands b and c were characteristic bands of fruits, leaves, and twig barks, respectively. The study suggests that the ADH isozyme in persimmon is organ-specific.
