摘 要:为揭示国内新型鸭肝炎病毒(DHV)基因组3′末端的序列特点,用3′RACE和RT-PCR克隆DHV分离株C-GY株的基因组3′末端序列,并以GenBank中已知全序列的22株DHV作为参照,进行比较分析。结果表明,C-GY株基因组3′末端包含1 359 nt的3D、终止密码子TAA3、66 nt的3′UTR和15 nt的poly(A)。与其他DHV相同之处:C-GY的3D蛋白含453个氨基酸,亦包含小RNA病毒RNA聚合酶的5个特征基序。C-GY的3′UTR长度与台湾新型DHV相同,仅比韩国新型缺少1个C,但比DHV血清1型(DHV-1)长52 nt。分析3D和3′UTR核苷酸序列,可见C-GY与16株DHV-1之间同源性为75%~77%和66%~68%、与2株台湾新型DHV之间为80%和91%、与4株韩国新型之间为96%~97%和96%~97%,表明C-GY与韩国新型DHV具有相近的遗传关系,C-GY株与本实验中参考DHV属于小RNA病毒科的同一个未命名的新属,而且C-GY属于DHV的基因C型,也说明新型DHV与DHV-1的基因组3′末端存在较高的序列变异性。The aim of this research was to analyze the 3′end sequence characteristics in genome of a new serotype of duck hepatitis virus (DHV). The authentic 3′end sequence in genome of Chinese isolate C-GY was obtained by using 3′RACE and RT-PCR techniques and compared with those of other DHV strains. The analysis showed that 3′end sequences in C-GY genome include 3D region of 1359 nucleotides (nt), the terminator codon TAA, 3′untranslated region (UTR) of 366 nt, and the poly (A) tail of 15 nt. Like other DHVs, the deduced 3D protein of 453 amino acids of C-GY contained the five motifs characteristic for RNA polymerase of picornaviruses. The 3′UTR of C-GY has the same length with that of the new serotype DHV isolated in Taiwan and contains a C deletion when compared to the new serotype DHV isolated in Korea. However, the 3′UTRs of C-GY is 52 nt longer than the 314 nt of DHV serotype 1 (DHV-1). The nucleotide sequences in 3D region and 3′UTR of C-GY are 75% -77% and 66% -68% identical to those of 16 DHV-1s, 80% and 91% identical to those of 2 Taiwan Residents new DHVs, and 96% - 97% and 96% - 97% identical to those of the four Korea new DHVs, respectively. The results revealed the closest genetic relationship between C-GY and Korean new DHVs and a large variation in the 3′ end sequence of genomes between new serotypes and DHV-1. Phylogenetic analysis indicated that DHV strain C-GY belong to the genotype C of DHV in the novel picomavirus genus proposed for DHV-1.
