2-氨基-6-甲基-3-乙氧羰基-5,7-二苯基-4,5,6,7-四氢吡啶并噻吩的合成与结构表征

摘　　要：首先利用醛酮缩合制得二烯酮(1),然后利用1与甲胺溶液作用制得哌啶酮(2),最后利用Gewald反应使2、氰乙酸乙酯与硫磺在吗啡啉催化下,得到未见文献报道的2-氨基四氢吡啶并噻吩衍生物3,产率60%～80%。所得产物的结构由元素分析,IR,1H NMR和MS所确证。Firstly, 1,5-diphenytpenta-1, 4-dien-3-one is synthesized using Claisen-Schmidt reaction. Then, 1-methyl- 2,6-diphenylpiperidin-4-one is synthesized using 1, 5- diphenylpenta -1, 4-dien-3-one and methylamine, At last, Gewald reaction was utilized to obtain ethyl 2 -amino -6-methyl -5 ,7-diphenyl -4, 5, 6, 7- tetrahydrothieno pyridine -3- carboxylate through the reaction of 1-methyl-2, 6-diphenylpiperidin-4-one with ethyl cynoacetate and sulfur in the presence of morpholine. The reaction condition and spectral properties were researched. The structures were comqrmed by elemental analysis , IR, ^1H NMR and MS.
