摘  要:目的探讨PTCH基因在肝细胞癌组织中的表达及其与肝癌病理分级的关系。方法提取手术切除18例不同病理分级的人肝癌、癌旁组织和6例胆管结石手术切除的正常肝组织中的总RNA并逆转录为cDNA。应用实时荧光定量RT-PCR法观察人肝细胞癌组织与癌旁组织中PTCH基因的相对表达水平,以及观察人正常肝组织中PTCH基因的表达水平。结果PTCH在所测18例肝细胞癌组织与癌旁组织中均有表达,在6例正常肝组织中不表达,且在病理分级为Ⅰ和Ⅱ级的癌组织中PTCH的mRNA表达水平显著高于癌旁组织(P<0.01),而在病例分级为Ⅲ级的癌组织中PTCH的mRNA表达水平显著低于癌旁组织(P=0.024)。结论随着肝癌恶性程度的增加PTCH的mRNA表达量呈递减趋势,提示PTCH可能参与了肝细胞的早期癌变过程。Objective The purpose of this study is to determine the expression level of PTCH in hepatocellular carcinoma (HCC) and the relationship between PTCH expression and the pathological grading of tumors, Method Total RNAs from 18 HCC tumor tissues and their matched adjacent non-malignant liver tissues were extracted and reverse-transcripted into cDNAs, Same method was used to prepare the cDNAs from 6 normal liver tissues from surgical resection specimens of patients with hepatic cholangiolithiasis. The expression levels of PTCH were determined by real time quantitative RT-PCR. Result Expression of PTCH was not detected in normal liver tissue. The expression level of PTCH in stage Ⅰ and stage Ⅱ tumor was higher than the normal adjacent nonmalignant tissues (P〈0.01). However, the expression level of PTCH in stage Ⅲ tumor was lower than the normal adjacent non-malignant tissues (P = 0.024). Conclusion The expression of PTCH mRNA was correlated with the histological differentiation and malignancy of HCC. The expression level was greatly decreased in poorly differentiated HCC. Our observation indicates that PTCH is involved in early stage of HCC development.
