摘 要:目的:探讨CD133在胃癌组织中的表达及其临床意义。方法:采用半定量逆转录聚合酶链反应(RT-PCR)检测38例手术切除的胃癌原发组织、癌旁正常胃粘膜组织和其中8例肝转移灶组织中CD133 mRNA的表达,免疫组织化学方法检测肿瘤组织中CD133蛋白的表达情况,分析CD133的表达与临床病理参数的关系。结果:胃癌癌旁、原发、转移组织中CD133 mRNA的灰度相对值分别为0.189±0.149、0.305±0.118和0.359±0.139,癌旁与原发组织之间差异有统计学意义(P<0.01),原发与转移组织之间差异无统计学意义(P>0.05)。免疫组化检测肿瘤组织中CD133蛋白表达,阳性细胞稀少。原发组织CD133 mRNA的表达与肿瘤大小和Ki-67表达相关(P<0.05),与性别、年龄、浸润深度、淋巴结转移分期、TNM分期、组织分化程度及p53表达均无关(P>0.05)。结论:CD133的表达与胃癌的发生有关,与转移的关系有待进一步研究,可能在胃癌的诊断中起一定的作用。Objective :To study the expression and clinical significance of cancer stem cell marker CD133 in gastric cancer tissues. Methods .. The expressiones of CD133 mRNAs which were extracted from human gastric cancer tissues(GC) and paracarcinoma gastric mucosa tissues(PGMT) of 38 patients were detected by RT-PCR, and the same way was used in tissues from metastasis of liver (ML) which were originated from 8 cases of total specimens. The protein expression of CD133 in tumor tissues was detected by immu- nohistochemisty(IHC). The relationships between CD133 and clinicopathological index were analysed. Results :The gray scale value of CD133 mRNA in PGMT, GC and ML was 0. 189±0. 149, 0. 305 ±0. 118 and 0. 359 ±0. 139, respectively. The expression of CD133 mRNA in GC was significantly higher than in PGMT(P 〈0. 01 ), but there was no significant difference in GC and ML(P 〉 0. 05). There were few positive cells expressing CD133 protein in tumor tissues which were stainned by by IHC. The expression of CD133 mR- NA in GC was correlated with tumor size and expression of Ki-67 (P 〈 0. 05 ) , there was no evidence indicating which was relative with sex, age, T stage, N stage, TNM stage, tumor differentiation and expression of p53 (P 〉 0. 05). Conclusion:The expression of cancer stem cell marker CD133 was related to tumorigenesis of gastric cancer, more research should be performed to discover the relationship between CD133 and neoplasm metastasis, CD133 can play an appropriate role in diagnosis of gastric cancer.
