摘  要:目的探讨川芎嗪联合顺铂对小鼠Lewis肺癌生长和转移的抑制作用及其机制。方法将40只接种Lewis肺癌的C57BL小鼠随机分成4组:对照组、顺铂组、川芎嗪组及联合用药组,每组10只,连续用药20天,于接种后第22天处死全部小鼠,剥离皮下肿瘤称瘤重并计算抑瘤率,取出双肺观察表面肿瘤转移情况,计算肿瘤肺转移发生率、肺表面转移结节数及肺表面结节转移抑制率,采用免疫组化检测肿瘤组织微血管密度(MVD)及P-选择素的表达水平。结果各用药组肿瘤的生长明显受到抑制,瘤重明显低于对照组(P<0.01),联合用药组抑瘤率明显高于顺铂组和川芎嗪组(P<0.05),各用药组肺表面转移结节数及转移发生率明显低于对照组(P<0.05或0.01);联合用药组肺表面结节转移抑制率明显高于顺铂组和川芎嗪组(P<0.05)。川芎嗪组、联合用药组MVD及P-选择素表达水平显著低于对照组(P<0.01)。结论川芎嗪可明显抑制Lewis肺癌细胞在小鼠体内的生长和转移,与顺铂联用有协同作用,其作用机制可能与抑制微血管生成和降低P-选择素表达有关。Objective To explore the inhibitory effect and mechanism of tetramethylpyrazine in combination with cisplatin on growth and metastasis of Lewis lung carcinoma in mice. Methods The Lewis lung carcinoma models of C57 BL lnice were established. Forty 3739 mice were randomly divided into 4 groups: control group( Group A), cisplatin group (Group B), tetramethylpyrazine group(Group C) and combination group (Group D). Every group had 10 mice. Different treatments were served from day 2 after transplantation and all mice were sacrificed after 22 days. The subcutaneous tumors were weighed to calculate inhibitory rate. The metastatic tumor foci on lung surface in mice were counted to calculate occmTence rate and inhibitory rate of metastases on lung surface. The expression of microvessel density (MVD) and P- selcctin were detected by immunhistochemical method. Results The tumor growth was suppressed significantly in cisplatin group,telramethylpyrazine group and combination group. The weights of tumor were markedly decreased in cisplatin group, tetramethylpymzine group and combination group colnpared with control group. The growth inhibition rate was decreased in combination group than cisplatin group and tetramethylpyrazine group. The metastatic foci and occurrence rate on lung surface in cisplatin group, tetramethylpyrazine group and COlnhination group were lower than control group. The inhibitory rate of metastases on lung surface was increased in combined group than cisplatin group and tetramethylpyrazine group. The expression levels of MCV and P- selectin were significantly lower in tetramethylpyrazine group and combination group than control group. Conclusion Tetramethylpyrazine has synergetic inhibitory effect with cisplatin on growth and metastasis of Lewis lung carcinoma cell in mice through inhibiting angiogenesis and decreasing expression of P - selectin.
