柔肝抑纤饮加用二至丸抗小鼠肝纤维化形成的对比研究

摘　　要：目的:考察柔肝抑纤饮加用二至丸后在抗小鼠肝纤维化形成方面与单用柔肝抑纤饮有无差别,明确加用二至丸后对原方有无增效作用,并阐明其机制。方法:建立小鼠血吸虫性肝纤维化模型,将动物随机分组:A组(正常对照组);C组(吡喹酮组);D组(柔肝抑纤饮组);E组(柔肝抑纤饮+二至丸组)。除A组外,各组分别灌服相应药物,检测ALT、ALB、PCⅢ、HA、肝组织HYp,行VG染色、免疫组化染色并测定肝组织TGF-β1、a-SMA、Ⅲ型胶原的表达。结果:E组、D组在降低ALT、HA、PCⅢ水平,减少Ⅲ型胶原表达等方面,均明显优于C组(P<0.05,P<0.01),但E组、D组之间比较未见显著性差异(P>0.05);E组在升高ALB,降低HYp,降低胶原染色(VG)百分比含量,以及下调TGF-β1、α-SMA的表达等方面优于D组(P<0.05)。结论:柔肝抑纤饮加用二至丸后对于抗小鼠肝纤维化的形成具有增效作用,值得今后深入研究。Objective ： Try to find the difference of the effects in preventing the form of liver fibrosis between Rougan Yixianyin Decoction only and the Decoction combined with Erzhi pill, thus reveal the mechanism of anti - fibrosis. Methods： The earlier stage of mice hepatic fibrosis model was made by infected with cercariae of schistosoma japonicum, 48 mice were divided into 4 groups： group A （Normal control group） , group C （Praziquantel group）, group D （ Rougan Yixianyin group） , group E （ Rougan Yixianyin ＋ Erzhi pill group）, mice of each group were infected with cercariae except group A. then, group D were fed with Rougan Yixianyin Decoction, group E with Rougan Yixianyin ＋ Erzhi pill, and group A,C were fed with physiological saline respectively for 40 days. Mice of each group were put into death on the 61 st days, ALT, ALB, HA, PC Ⅲ in serum and HYp of liver tissue were tested ; The histopathology of hepatic tissue was observed too （ by HE/VG dye） ; The expression of TGF - β1 , α - SMA and collagen type Ⅲ were tested by quantitative a- nalysis. Results：Group D and E were much better than Group C in decreasing ALT,HA, resisting the expression of collagen type Ⅲ （P 〈 0.01 ） ; In terms of increasing ALB, decreasing HYp, resisting the expression of TGF - β1,α -SMA, Group E was much better than Group D （ P 〈 0.05 ）. Conclusions ： The anti - fibrosis effects of Rougan Yixianyin plus Erzhi - pill were proved to be much better than using Rougan yixianyin only.
