摘  要:目的观察降钙素对人骨巨细胞瘤(giant cell tumor of bone,GCT)组织中纯化的破骨细胞的数量以及细胞核因子-κB受体活化因子配基(RANKL)蛋白表达的影响。方法利用破骨细胞贴壁快以及耐胰蛋白酶的特性,采用0.25%胰蛋白酶和0.2%I型胶原酶来分离纯化骨巨细胞瘤中的破骨细胞,设立对照组和试验组,在实验组培养液中加入浓度为10-8mol/L的降钙素,以抗酒石酸酸性磷酸酶(TRAP)染色观察破骨细胞数目、形态,用免疫组化方法观察RANKL的表达。结果从骨巨细胞瘤中纯化方法得到的破骨细胞数量较多,加药组破骨细胞数目较对照组明显减少(P<0.05)。RANKL的蛋白表达两组未见明显差异(P>0.05)。结论降钙素可显著的抑制骨巨细胞瘤中纯化破骨细胞的分化和功能。Objective To observe the effects of calcitonin on the number and expressions of receptor activator of NF--KB ligand (RANKL) in osteoclasts puried from giant cell tumor of bone (GCT) . Methods Osteoclasts and their precursors were isolated from giant cell tumor of bone (used by 0.25% trypsin and 0.2% collagenase type I ). Osteoclasts were cultured in a - MEM and 10-8 mol/L of calcitonin added into the experimental group. After 2 days,the number, morphology of the osteoclasts were detected with tartrate - resistance acid phosphatase (TRAP) staining. Immunohistochemistry was used to examine the expressions of RANKL in osteoclast. Results The Number of osteoclasts was decreased in the experimental group ( P 〈 0.05 ) but there was no difference for RANKL proteinum between two groups. Conclusion Calcitonln couid inhibit differentiation and function of osteoclast purled from giant cell tumor of bone.
