摘 要:目的:含双螺旋环的Id1(inhibitor of DNA binding or inhibitor of differentiation),在器官组织中的表达具有组织特异性。近年研究显示其高表达与某些肿瘤的发生、发展密切相关。本研究检测了肾正常组织、肾良性囊肿和肾癌中Id1蛋白的表达,并分析其表达量与肾癌细胞分化类型的相关性。方法:用免疫组化SP法检测10例肾癌旁正常组织、38例肾细胞癌和10例肾良性囊肿组织中Id1的表达。用JD801形态图像分析系统对图像进行定量分析。结果:Id1蛋白在正常肾组织中在肾小管上皮细胞内高表达;在肾良性囊肿组织中表达量明显减少;在肾癌细胞中均阳性表达(38/38)。颗粒细胞癌中Id1的表达水平最高,其次为混合细胞癌,透明细胞癌表达量较少。结论:人正常肾小管上皮细胞内高表达Id1;良性囊肿组织中Id1表达减少;肾癌组织中Id1蛋白有广泛表达,且分化差的癌细胞类型表达量高,表明Id1与肾癌细胞的恶性度和预后密切相关。Objective: The helix - loop - helix protein Id - 1 ( inhibitor of DNA binding or inhibitor of differentiation) serves to prevent basic helix -loop -helix transcription factors from binding to DNA, thus, inhibiting the transcription of differentiation associated genes. The expression of Id1 has tissue specificity. Over expression of Id - 1 has been reported in certain tumors. This study aimed to detect the expression of Id1 protein in human renal, renal cyst and renal cell carcinoma ( hRCC ) ; to analyse the relationship between Id1 protein and renal cell differentiation. Methods:Formalin -fixed, paraffin embedded sections of 38 cases of hRCC, 10 cases of normal tissues and 10 cases of renal cyst were detected by SP immunohistochemistry. Figures were further analyzed by JD801 software. Results:In normal renal tissues, high positive immunostaining of Id - 1 was only found in the cells of renal tubule. There were no positive staining in glomeruli and interstitium. Decreased expression was observed in renal cyst. In contrast, all of renal cancer tissues included glomeruli and interstitium showed moderate to high positive Id1 expression. The highest expression of Id1 was observed in granular cell carcinoma of kidney. The second was in mixed renal cell carcinoma. The lowest was in clear- cell carcinoma. Conclusion:There is high expression of Id1 in normal renal tubule. Lower expression of Id1 is in renal cyst. All tissues of hRCC show positive expression of Id1. Id1 expression correlates with the differentiated types of hRCC, suggesting that Id1 associates with renal tumor malignancy and progression.
