摘  要:本文运用了三种不同的盐析法 ,四种不同的有机溶剂提取法进行试验 ,对比摸索出一种方法简单 ,活性高 (达 130× 10 4 u/ g± 6× 10 4 u/ g) ,稳定性好 ,适用于工业生产应用的木瓜蛋白酶纯化工艺。其方法是 :取新鲜木瓜乳汁 ,加适量偏重亚硫酸钠 ,搅匀 ,用 4 0 % NH4 OH调 p H至 9.0搅拌后抽滤 ,滤液加冰冻无水乙醇沉淀 ,乙醇浓度为 6 0 % ,低温反应 30 min,离心取沉淀物 ,用无水乙醇 ,丙酮脱水 ,真空干燥 ,充氮封存。Comparison was made among 3 salt out methods and 4 orgainc solvent extraction ones in this article. One of them was accepted as a simple and stable method suitable for purifying papain with high activty (130×10 4u/g±6×10 4u/g) in industrial manufacturing. The process is illustrated as follows: Fresh Latex of Carica Papaya was extracted and stirred with appropriate amount of sodium metasulfite. After its pH value was adjusted to 9.0 with 40% NH 4OH,the mixture was filtered by suction and its liquid part was precipitated with cold absolute alcohol at the saturation level of 60%. The reaction took 30 minutes. Then the precipitate was obtained by centrifugation and dehydrated with absolute alcohol and acetone. At last the product was packed and stored by exhausting and nitrogen filling.
