摘 要:用紫外线、硫酸二乙酯(DES)、He-Ne激光诱变和UV+DES复合诱变,对1株产碱性纤维素酶的软化芽孢杆菌IS-B进行诱变育种,获得产酶能力是出发菌株5倍且产酶性能稳定的高产菌株IS-B4。采用单因素分析和正交试验对其发酵培养基进行优化,优化后培养基配方:蔗糖2%,酵母膏2%,NaCl 0.5%,KH_2PO_4 0.1%。在温度36℃,pH 9.0,接种量5%,250 mL三角瓶装液量为60 mL的发酵条件下,酶活力可达56.0 U/ mL。The original strain Bacillus macerans IS-B was mutated by UV,DES, He-Ne laser and UV+DES and its enzyme productivity was 5 fold higher than that of parent strain. The continuous 10 generations test shows that the mutant IS-B4 is stable in the alkaline cellulase (CMCase) production. The optimal fermentation conditions for CMCase production were determined by a single-factorial experiment and orthogonal experiment. The results showed that the optimal conditions were as follows, saccharose 2 %, yeast extract 2%, NaCl 0.5%, KHaPO4 0.1%. The incubation temperature 36℃, inoculum amount 5%, pH 9.0, the load of 250mL-flask was 60mL and the enzyme activity could reach to 56.0 U/mL.
