肾透明细胞癌中转化生长因子-β1、基质金属蛋白酶2表达及其临床意义的相关性研究

摘　　要：目的:研究转化生长因子-β1(TGF-β1)、基质金属蛋白酶2(matrix metalloproteinases 2,MMP2)在肾透明细胞癌(renal clear cell carcinoma,RCCC)中的表达及临床意义。方法:采用免疫组织化学SP法测定80例RCCC及30例癌周组织中TGF-β1与MMP2的表达,同时采用逆转录-多聚酶链反应(RT-PCR)测定43例RCCC及43例癌周组织中TGF-β1与MMP2基因的表达。结果:RCCC中TGF-β1、MMP2表达强度均明显高于癌周组织,TGF-β1、MMP2表达与RCCC病理分级、临床分期相关;RCCC中TGF-β1与MMP2表达强度具有相关性;单因素生存分析显示TGF-β1、MMP2高表达与患者预后不良有关(P均<0.05)。结论:TGF-β1、MMP2基因的表达参与了RCCC的发展,并且二者之间具有一定的相关性,提示可以作为判断RCCC预后的二个有效的联合分子指标。Objective：To investigate the expression of transforming growth factor beta 1 （TGF-β1） and matrix metalloproteinases 2 （MMP2） in human renal clear cell carcinoma （RCCC） and their clinical significance. Methods：The protein expression of TGF-β1 and MMP2 in 80 cases of RCCC and 30 cases of tissues adjacent to cancer were determined with immunobistocbemical SP method. The mRNA expression of TGF-β1 and MMP2 gene in 43 cases of RCCC and 43 cases of tissues adjacent to cancer were determined with reverse transcription-polymerase chain reaction（RT-PCR）. Results：In RCCC group TGF-β1 and MMP2 expression intensity was obviously higher than that in tissues adjacent to cancer. The expression of TGF-β1 and MMP2 were correlated with the pathologic grade and clinical stage of RCCC. There was a statistically significant correlation between expressions of TGF-β1 and MMP2. The high expressions of TGF-β1 and MMP2 significantly correlated with the poor prognosis of patients with RCCC by univariate survival analysis （P 〈 0.05, respectively）. Conclusion：Up-regulation of TGF-β1 and MMP2 expression may be involved in the progression of RCCC. The correlation was found between TGF-β1 and MMP2. This indicates that TGF-β1 and MMP2 can be used as a useful associatedmolecular marker for prognosis of RCCC.
