β-七叶皂苷钠对大鼠脑出血模型脑水肿治疗作用的超微结构研究  被引量:6

摘  要:目的:研究β-七叶皂苷钠对大鼠脑出血后脑水肿的影响。方法:用立体定位法采用新鲜自体血注入大鼠尾状核建立脑出血模型,将实验大鼠随机分为治疗组和对照组;治疗组分别于术后1 h、3 h、6 h和24 h经尾静脉注射七叶皂苷钠(2.5 mg/kg);对照组于相应时间点尾静脉注射生理盐水。治疗组连续用药3天(1次/天),对照组连续注射生理盐水(1次/天)。评估大鼠治疗前及治疗后的行为学改变、测定脑含水量和电镜下检查血肿周边脑组织细胞的超微结构改变。结果:治疗组脑含水量明显低于对照组(P<0.05),各治疗组间相比差异无显著性(P>0.05)。用药3天后治疗组与对照组相比行为学评分有统计学意义(P<0.05);治疗组各组间比较无显著性差异(P>0.05)。电镜观察显示治疗组血肿周围脑组织内星形细胞足突水肿较生理盐水对照组有所减轻;4个治疗组之间相比,出血后1h治疗组脑水肿的效果最佳。结论:β-七叶皂苷钠对脑出血后早期脑水肿有一定的治疗作用。Objective:To observe the effect of β-aescinate administerd in different time on changes of edema after intracerebral hemorrhage in rats.Methods:The intracerebral hemorrhage animal models were stablished by stereotaxical injecting autologous blood. Rats were randomly divided into treated group and control group. After operation, β-aesclnate or normal saline were injected into the caudal vein of 1 h, 3h, 6h and 24h treated group or control group respectively. The water content of perihematoma tissue, evaluation of mouse ethology and ultrastructural changes of pefihematoma tissue were investigated. Results:Water content in the perihematoma tissue and ethology score of rats were lower in treated group than that in the control group when rats were treated 3 days( P 〈 0.05). However, there were no statistic significance between 4 subgroups( P 〉 0.05); Compared with the control group, pyknotic neuron, edema astrocytes and foot processes of astrocytes with depressed capillary wall were milder than in the treated group. In control group some large vacuoles and devoid of organelles were seen in the cytoplasm, while in treated group of 1 h after intracerebral hemorrhage, the uhrastructure of the neuron was apparently normal in appearance. Conclusion: β-aescinate has an evidently protective effect on cerebral edema after intracerebral hemorrhage in rats.
