摘 要:为了筛选麻痹性贝毒毒性早期预测指标,采用膝沟藻毒素GTX-1,4对小鼠进行暴露,剂量为1μg/kg,时间为60、901、20 min.结果发现暴露组脑中乙酰胆碱(Ach)含量在60 min明显高于空白组(P<0.01),90 min时显著低于空白组(P<0.05);乙酰胆碱酯酶(AchE)活力在90 min明显低于空白组(P<0.01);NO含量在3个时间节点上均没有明显变化;一氧化氮合酶(NOS)活力随时间延长呈下降趋势,120 min时NOS活力低于对照组,差异具有统计学显著性(P<0.01).从而推测:神经递质参与了麻痹性贝毒对脑损害的毒性过程;NOS是麻痹性贝毒毒性相对敏感的指标.To probe into the prelude marker of toxicity of paralytic shellfish poisoning (PSP), the mice were exposed to 1 μg/kg toxin GTX-1,4, the sampling times were 60, 90, 120 min respectively. The contents of ACh,NO and the activities of ACHE, NOS in mice brain were measured. The results show the content of ACh increases significantly in the brain of acute exposure mice at 60min , compared with control group, (P〈0.01), but at 90 rain it is lower (P〈0.05). The activity of AChE is lower than that of control group at 90 min (P〈0.01). There is no changes of NO content in exposure mice compared with control group. The activity of NOS is downtrend at 60,90,120 min respectively, and lower significantly than that of control group at 120 min(P〈0.01). It can conclude neural transmitters participate in the toxicity process of brain injury by PSP, the NOS activity in mice brain might be the bio-index of PSP.
