摘 要:目的探讨华佗健聪汤对β淀粉样多肽(Aβ)所致神经毒性的拮抗作用。方法以原代培养的SD乳鼠神经元为材料,随机分为正常组、模型组、华佗健聪汤组、人参皂苷Rg1对照组。除正常组外,其余各组用Aβ1-42造成神经元损伤,华佗健聪汤组用华佗健聪汤给SD成年大鼠灌胃(含生药20 g/kg)后获取的大鼠含药血清进行干预,人参皂苷Rg1对照组用人参皂苷Rg1进行干预。观察并拍摄细胞形态,检测MTT值,取细胞上清夜检测LDH、T-AOC、NO等指标。结果华佗健聪汤组与模型组相比,MTT、T-AOC显著增高,差异具有统计学意义(P<0.05);而LDH和NO则明显降低,差异具有统计学意义(P<0.05)。结论华佗健聪汤对Aβ所致神经元损伤的大鼠有较好的保护作用,抗自由基的氧化损伤作用可能是其保护神经元的作用机制之一。Objective To study the function of Huatuo Jiancong decoction on antagonising Aβ toxicity to neuron. Method The primary cultured murine neurons were randomly divided in-to control group, model group, Ginsenoside Rg1 group and Huatuo Jiancong decoction group. All the rats, beside the rats in control group, were damaged by Aβ1-42. In Huatuo Jiancong decoction group, the SD rats were treated by the decoction and then thier serum containing medicine ingredients was taken for antagonising Aβ toxicity, Ginsenoside Rgl in the control group was used for that. Then, the photographs of cell shapes were observed and MTT and calculating the living rate detected; the LDH, T-AOC, NO in supernatant liquid were detected. Results MTT and T-AOC were increased in Huatuo Jiancong decoction group, compared with model group (P〈0.05), while LDH and total NO decreased (P〈0.05). Conclusion The decoction can protect remarkably neuron poisoned by Aβ, and the mechanism may be its antioxidation function.
