CD147蛋白表达与子宫颈癌生物学行为的关系

摘　　要：目的:检测CD147蛋白、MMP2 mRNA在子宫颈癌中的表达,探讨它们与子宫颈癌浸润、转移的关系。方法:采用免疫组织化学Maxvison法、RT-PCR技术检测65例子宫颈癌、21例癌旁正常组织中CD147蛋白、MMP2 mRNA的表达。应用图像分析软件Image-Pro Plus6.0及Image Tool分别对CD147和MMP2 mRNA的表达行平均光密度和灰度分析。结果:宫颈癌组织中CD147蛋白和MMP2 mRNA的表达高于癌旁正常组织,二者之间差异具有显著性(P<0.01),宫颈癌组织中CD147蛋白和MMP2 mRNA的表达与组织学分级、TNM分期及淋巴结转移密切相关(P<0.05),而与年龄、肿块大小、组织学类型无相关性(P>0.05)。本组中CD147蛋白和MMP2 mRNA的表达存在正相关(r=0.513,P<0.05)。结论:CD147可能促进了MMP2的表达,二者可成为判断宫颈癌生物学行为和指导临床治疗的指标。Objective： To detect the expression of CD147 protein and MMP2 mRNA in cervical carcinoma tissues and further to explore the correlation between their expression and the cervical carcinomas the invasion and meatastasis. Methods： Immunohistochemistry named Maxvison and RT - PCR were used to respectively detect the expression of CD147 and MMP2 mRNA in both 65 cases of cervical carcinoma and 21 cases of pericancerous tissues and quantitative analysis of mean density of the expression of CD147 and grey of MMP2 mRNA were conducted respectively with the image analytic software： Image- Pro Plus （ version 6. 0 ） and Image Tool Results： In comparison with corresponding pericancerous tissues group, the expression of CD147 and MMP2 mRNA in cervical carcinoma group was significantly higher （P 〈 0. 01 ）. The expression of CD147 and MMP2 mRNA were correlated with histological grade, TNM stage and lymph node metastasis （P 〈 0. 05） and not correlated with age, lump and histological type. The expression of CD147 was positively correlated with the expression of MMP2 mRNA in cervical carcinoma （r = 0. 513, P 〈 0. 05 ） . Conclusion： The expression of CD147 was positively related to that of MMP2 mRNA, which means CD147 may promotes the expression of MMP2 and both of them jointly may be a reliable indicator to determine biological behaviors and clinical therapy of cervical carcinoma.
