摘 要:目的研究ClC-3氯通道对鼻咽癌细胞CNE-2Z周期的影响。方法利用反义技术抑制细胞内ClC-3的表达,用台盼蓝拒染法检测细胞存活率,MTT法检测细胞增值能力,流式细胞仪测定细胞周期分布。结果ClC-3反义寡核苷酸剂量依赖性抑制鼻咽癌细胞的增值,胞内ClC-3的表达下调使细胞周期进程明显受到影响,细胞停滞于G1期,而ClC-3反义寡核苷酸对细胞存活率没有影响。结论CNE-2Z细胞内ClC-3的表达下调可阻滞细胞于G1期而使细胞增值受到抑制。提示氯通道ClC-3参与了鼻咽癌细胞CNE-2Z的细胞周期调控。Objective To study effects of ClC-3 on the cell cycle of human nasopharyngeal carcinoma cell strain CNE-2Z.Methods Antisense technology was used to inhibit the expression of endogenous ClC-3.MTT assay was applied to detect cell proliferation.Cell cycle distribution was analyzed by the flow cytometry.Cell viability was determined by the trypan blue assay.The expression levels of ClC-3 mRNA was detected by RT-PCR.Results The expression of ClC-3 mRNA was down-regulated in CNE-2Z cells treated with ClC-3 antisense oligonucleotides which suppressed CNE-2Z cell proliferation in a dose-dependent manner.ClC-3 antisense oligonucleotides also arrest CNE-2Z cells in G1 phase of cell cycle.,but no obvious change in the survival rate of CNE-2Z.Conclusion ClC-3 antisense oligonucleotides suppressed cell proliferation by arresting cell in G1 phase.The results suggest that ClC3 plays an important role in the regulation of cell cycle.
